摘 要:目的:探讨KATP通道的功能改变与胶质瘤细胞增殖的关系。方法:鉴定KATP通道在胶质瘤细胞上的表达,并比较对照组、KATP通道激动剂、KATP通道阻断剂影响胶质瘤细胞的增殖情况。结果:证实KATP通道在胶质瘤细胞中呈高丰度表达,KATP通道的激动剂二氮嗪(100μmol/L)显著促进细胞增殖。KATP通道的阻断剂甲糖宁(100μmol/L)显著性抑制细胞增殖。两者对细胞增殖的影响均呈浓度依赖方式。结论:KATP通道的确参与胶质瘤细胞增殖过程的调节,为胶质瘤的治疗提供新的靶标。Objective To study the effect of ATP-sensitive potassium channels activity on glioma cells proliferation. Methods Tested KATP channels expression in glioma cells, and to examined glioma cells proliferation in control group, group treated with KATP channels antagonist and group treated with KATP channels agonist. Results The KATP channels were highly expressed in glioma cells. Diazoxide ( 100 μmol/L), the KATP channels agonist promoted the glioma cells proliferation. In contrast, tolbutamide (100 μmol/L), the KATP channels antagonist which blocked the cellular sulphonylureas receptor, significantly inhibited the glioma cells proliferation. And the effects of them on glioma cells proliferation were dosage-dependent. Conclusions KATP channels are important in pathological cell proliferation and open a promising pathway for novel targeted therapy.
