摘  要:目的研究中国汉族重性抑郁症(MDD)患者5-羟色胺2A受体基因多态性与帕罗西汀抗抑郁临床疗效之间的关系。方法 117例抑郁症患者给予帕罗西汀(20~50mg.d-1)治疗8周,用汉密尔顿抑郁量表(HAMD-17)评估临床疗效,聚合酶链式反应(PCR)和限制酶切的方法检测A1438G基因型。结果有效率、缓解率及不良反应发生率在A1438G不同基因型及等位基因间分布差异无统计学意义(P>0.05);G等位基因携带者睡眠症状改善好。结论 A1438G多态性与帕罗西汀对睡眠症状的改善有关。Objective To investigate the relationship between serotonin receptor 2A(5 -HT2A) gene polymorphism and clinical response to paroxetine in Chinese major depressive disorder (MDD) patients. Methods One hundred senventeen patients affected by MDD were treated with paroxetine for 8 weeks. The clinical response was evaluated with the Hamilton Rating Scale for Depression ( HAMD - 17 ) , and adverse drug reactions were assessed at each visit. PCR - RFLP was used to detect A1438G gene polymorphism. The relations between genetypes and clinical response ,adverse drug reactions were analysed. Results There was significant differences of main clinical response and adverse drug reactions (ADRs) among different genotypes and allele carriers. There were significant differences between G carriers and G non - carriers in sleep symptom improvement. Conclusion The 5 - HT2A reeeptor gene polymophism A1438G was associated with the sleep symptom improvement of MDD.
