摘  要:目的:探讨脂肪酸合酶抑制剂对人结肠癌细胞增殖的影响。方法:应用细胞形态学、MTT法及成集落实验方法进行检测和观察。结果:癌细胞在脂肪酸合酶抑制剂浅蓝菌素作用下,细胞增殖被阻遏,并呈现剂量—效应关系。同时发现癌细胞呈现细胞凋亡的形态学改变。凋亡细胞表现为细胞固缩,核染色质凝聚、边集或断裂。浅蓝菌素对人成纤维细胞增殖无明显影响。结论:脂肪酸合酶抑制剂可能通过抑制LoVo细胞内源性脂肪酸合成,诱发细胞凋亡来阻遏LoVo细胞的增殖。bjective: The purpose of the experiment was to investigate the effects of fatty acid synthase inhibitor on the proliferation and apoptosis of human colonic cancer cells. Methods: Cell morphology, microculture tetrazolium assay (MTT) and colonyforming test were used for detection of fatty acid synthase inhibitor cerulenin which inhibittes proliferation and induces apoptosis in human colonic cancer cells (LoVo). Results: Cerulenin inhibited the proliferation of LoVo cells in vitro and showed a doseresponse relationship.Cerulenin also induced death of LoVo cells via apoptosis. The demonstration of apoptotic cells included according to the cell shringkage, condensation and fragmentation of chromosomes. Conclusion: The fatty acidsynthase inhibitor could suppress the growth of human colonic cancer cells and induced their death via apoptosis by the inhibition of endogenously synthesized fatty acids in cancer cells.
