摘 要:目的探讨褪黑素(MT)对Balb/c小鼠胰腺癌皮下移植瘤的治疗作用及对其自然杀伤(NK)细胞活性的影响。方法在Balb/c小鼠皮下接种胰腺癌SW1990细胞建立荷瘤鼠模型,每组分别用生理盐水、MT低剂量(10mg·㎏-1·d-1)、MT高剂量(20mg·㎏-1·d-1)干预,定期测量肿瘤大小,绘制肿瘤生长曲线,称瘤质量,并用MTT法检测小鼠脾脏(NK)细胞活性。结果与对照组肿瘤体积[(1.476±0.075)cm3]比较,MT低剂量组[(0.998±0.112)cm3]、MT高剂量组[(0.756±0.128)cm3]肿瘤体积显著减小(P<0.01),并且MT高剂量组肿瘤体积明显小于低剂量组(P<0.05)。与对照组肿瘤质量[(1.537±0.106])g]比较,MT低剂量组[(0.898±0.125])g]、MT高剂量组[(0.636±0.081)g]瘤质量显著减轻(P<0.01),并且MT高剂量组瘤质量明显轻于低剂量组(P<0.05)。与对照组NK细胞的杀伤活性[(18.07±1.23)%]相比,MT低剂量组[(44.27±3.19)%]、MT高剂量组[(45.16±3.20)%]NK细胞的杀伤活性显著增强(P<0.01),MT低剂量组与MT高剂量组之间无明显差异(P>0.05)。结论 MT能抑制小鼠皮下移植性胰腺癌的生长,增强荷瘤鼠的免疫功能。Objective To investigate the effect of melatonin for the treatment of pancreatic cancer in Balb/c mice subcutaneous xenograft model and effect of NK cell activity in mice.Methods Pancreatic cancer model was established by subcutaneously inoculating SW1990 cells into the right flank in Balb/c mice.Tumor size was measured regularly after injection of saline,low dose of MT(10 mg·㎏ -1·d -1),high dose of MT(20 mg·㎏ -1·d -1).Results Compared with the controls [(1.476 ± 0.075) cm3],tumor volumes in low dose MT group[(0.998 ± 0.112) cm3]and high dose MT group[(0.756 ± 0.128) cm3],were significantly shrunk(P 0.01).The significant difference in weight of tumor in low dose MT group[(0.898 ± 0.125]) g]and high dose MT group [(0.636 ± 0.081) g]was found both of which was significantly lowered than that in the controls[(1.537 ± 0.106]) g](P 0.01).Tumor volumes and weight in the high dose MT group was significantly different with that in the low dose MT group(P 0.05).The killing activity of NK cell significantly increased in the low dose MT group [(44.27 ± 3.19) % ]and the high dose MT group [(45.16 ± 3.20) % ]than that in the controls [(18.07 ± 1.23) % ](P 0.01).Meanwhile,there was no significant difference between the MT low-dose group and the MT high-dose group(P 0.05).Conclusion MT could inhibit the growth of tumor and improve immune function in mice.
