生长抑素对胃癌SGC-7901细胞VEGF、NF_κB表达的影响

摘　　要：目的研究生长抑素对胃癌SGC-7901细胞血管内皮生长因子(VEGF)、核转录因子-κB(NFκB)表达的影响。方法不同浓度生长抑素处理SGC-7901。MTT法测量吸光度值,计算细胞生长抑制率;免疫细胞化学(SP)法检测VEGF、NFκB蛋白表达的变化;RT-PCR半定量检测VEGF、NFκB mRNA表达水平。结果实验组与对照组比较,细胞生长抑制率有明显差异(P<0.05),同一实验组不同时间点(24、48、72h)间比较,细胞生长抑制率也有明显差异(P<0.05),表明生长抑素对SGC-7901细胞生长抑制作用有剂量时间依赖性;不同浓度生长抑素处理48h后,SGC-7901细胞VEGF、NFκB蛋白表达下降,实验组与对照组比较有明显差异(P<0.05);mRNA的表达呈浓度依赖性降低,实验组与对照组比较有明显差异(P<0.05)。结论生长抑素能下调胃癌SGC-7901细胞VEGF、NFκB表达,抑制胃癌细胞增殖。Objective To study the effect by Somatostain on the expression of vascular endothelial groth factor（VEGF）,nuclear factor kappa B（NFκB） in human gastric cancer cell line SGC-7901.Methods Exponential phase human gastric cancer cell line SGC-7901 was treated by Somatostain in various concentrations in vitro.By means of MTT assay,the absorbance value was detected and inhibition rate was calculate.The expression of VEGF,NFκB protein was analyzed by immunocytochemistry（SP）.The expression of VEGF,NFκB mRNA was determined by RT-PCR.Results The difference in inhibitory rate between the experiment group and the control group was significant（P 0.05）.It was also significant among the different time groups（24,48,72 h）（P 0.05）.It showed that the inhibitory effect was doseand-time dependent.After 48 h,the expression of VEGF and NFκB protein was down regulated.The difference between the experiment group and the control group was significant（P 0.05）.The expression of VEGF and NFκB nRNA was down regulated through ladder strap as the increasing of Somatostain concentration.The difference between the experiment group and the control group was significant（P 0.05）.Conclusion Somatostain can down regulate the expression of VEGF and NFκB,and it also can control the growth and apoptosis of the human gastric cancer cell line SGC-7901.
