摘 要:目的探讨氯化钴(CoCl2)对原代培养的猪肺动脉内皮细胞钙离子浓度的影响。方法原代培养猪肺动脉内皮细胞,分别用不同浓度的CoCl2刺激2min,用双波长荧光分光光度计观察细胞质内钙离子浓度的变化;另外,用1mmol/L的CoCl2刺激细胞不同时间,观察细胞质内钙离子浓度的变化。结果不同浓度的CoCl2对猪肺动脉内皮细胞内钙离子浓度无显著影响;1mmol/L的CoCl2刺激细胞不同时间也对细胞内钙离子浓度无显著影响。结论与物理缺氧方法不同,化学缺氧试剂CoCl2对原代培养的猪肺动脉内皮细胞的钙离子浓度没有明显影响,表明猪肺动脉内皮细胞对CoCl2的刺激有一定的耐受性。Objective To study the role of Cobalt Chloride (CoCl2) on the cytosolic calcium concentration of primary cultured porcine pulmonary artery endothelial cells.Methods The primary cultured porcine pulmonary artery endothelial cells were treated with different concentration of CoCl2 for 2 minutes,and the changes of cytosolic calcium concentration were observed by dual-wavelength fluorescence spectrophotometer.We also observed the changes of cytosolic calcium concentration after stimulating the cells with CoCl2 at the dosage of 1mmol/L for different time.Results CoCl2 at different concentration could not influence cytosolic calcium concentration of primary cultured porcine pulmonary artery endothelial cells.And cytosolic calcium concentration did not change after the cells were stimulated with CoCl2 at the dosage of 1mmol/L CoCl2 for different time.Conclusion Different from physical hypoxia,CoCl2 could not alter the cytosolic calcium concentration of primary cultured porcine pulmonary artery endothelial cells which may tolerate the stimulus of CoCl2 in a certain degree.
