摘  要:目的:观察伯氏疟原虫(P.berghei)小亚单位核糖体RNA(SSUrRNA)在斯氏按蚊体内的转录类型和动态。方法:应用RT-PCR方法,特异地扩增伯氏疟原虫A型和S型SSUrRNA,检测斯氏按蚊虫体内S型SSUrRNA的转录水平。结果:吸感染血的斯氏按蚊体内仅见S型SSUrRNA的条带。整体检测从第2天开始查见,第8天明显增强,第12~16天达到高峰。在蚊腹部S型SSUrRNA转录动态与整体检测结果相似。胸部于第12天查见该基因的转录。结论:斯氏按蚊体内伯氏疟原虫仅转录S型SSUrRNA,它的转录动态与疟原虫在蚊体内的发育密切相关。Objective: To detect and observe types and transcription dynamics of small subunit ribosomal RNA (SSUrRNA) of Plasmodium berghei in Anopheles stephensi. Methods: The A and Stype SSU rRNA of the parasites were distinctively amplyfied by RTPCR. The transcription levels of the StypeSSUrRNA in different parts of the mosquitoes were detected. Results: Only the Stype SSUrRNA was detected in the parasiteinfected mosquitoes. The Stype SSUrRNA electrophoresis band was seen on second day after engorgment of the mosquitoes. On d 8, its intensity increased remarkably and reached the peak on d 1216. The transcription dynamics of the Stype SSUrRNA in the mosquito abdomen were similar to those of the whole mosquitoes. In contrast, the transcription of the SSUrRNA was detected in the mosquito chest on d 12. Conclusion: Only the Stype SSUrRNA of the P.berghei is transcribed during their sporogony development.
