摘  要:建立高分辨率的套式PCR-SSP,对17个HLA-A*02等位基因分型。利用国际标准分型细胞进行特异性鉴定,并在54例A*02阳性的无血缘关系汉族健康人中应用。共检测到5个A*02亚型,显示出较高多态性:其中A*0211(n=2)首次在东方人群检测到;A*0201频率最高,占有48.2%;其余依次为A*0206、A*0207和A*0203。套式PCR-SSP既适用于移植供体选择,又可用于人类进化研究。A nested PCRSSP was developed for the determination the 17 known HLAA*02 alleles. The specificity of the reaction was tested by a panel of international standard genomic DNAs. The method was applied to 54 nonbloodrelated healthy individuals recruited from Chinese Han population positive for A*02 alleles. Five A*02 subtypes were found indicating there was, relative polymorphism among the Han individuals. A*0211 (3.7%), which occurred at a low frequency of 1.3% in the Caucasians, was detected in the Orient for the first time. A*0201 ranked first in the incidence (48.2%). A high proportion of A*0206 A*0203 and A*0203 were also found. The nested PCRSSP could be used as a tool both for the study of human evolution and for HLA matching before tissue or organ transplantation, especially unrelated donorrecipient transplantation.
