摘　　要：目的制备兔抗人S100A7(hS100A7)多克隆抗体。方法制备原核表达重组人S100A7,用纯化的hs100A7为免疫原免疫新西兰大白兔,制备兔抗人S100A7抗体,并以间接ELISA测定抗体效价,以Western blot和免疫组织化学法鉴定抗体的特异性。结果成功制备了重组人S100A7及其抗体,ELISA结果显示抗体效价达1∶16000,Western blot和组织化学法分析表明该抗体能特异结合hS100A7。结论以纯化的重组人S100A7为免疫原,成功地制备了效价高、特异性强的兔抗人S100A7抗体。Objective To prepare antibody against human S100A7 （hS100AT）.Methods The recombinant hS100A7 was prepared by prokaryotie expression and was used to immunize the rabbit. The titer and the specificity of the rabbit anti-hS10OA7 antibody were analyzed by indirect ELISA and Western blot respectively. Results The recombinant hS100A7 and the antibody against hS100A7 was successfully prepared, the titer of the generated antibody was 1：16000 by ELISA. The results of Western blot and immunohistochemistry showed that this antibody not only could bind with recombinant hS10OA7 but also native hS100A7 specifically. Conclusion The rabbit anti-hS10OA7 antibody with high titer and high specificity has been prepared by using purified recombinant hS100A7 as immunogen.
