摘 要:研究了赭曲霉对16α,17α-环氧黄体酮(EP)的C11α-羟基化工艺条件。考察了接种量、摇床转速、底物投料方式及投料时间等工艺参数对转化率的影响,初步确定最佳转化工艺条件如下:转化培养基初始pH值为5.0、接种量为2.0%、摇床转速为200 r.min-1、28 h时投加乙醇溶解的EP、EP浓度为10 g.L-1,此条件下转化率达80.5%。将羟丙基-β-环糊精(HP-β-CD)应用于转化反应中,在转化反应进行4 h时添加与EP摩尔比为1.25∶1的HP-β-CD,转化率达到93.1%,较未添加HP-β-CD的转化率提高了12.6%。The C11α-hydroxylation of 16α,17α-epoxyprogesterone(EP) by Aspergillus ochraceus 405 was studied.The most important fermentation factors,such as inoculum amount,rotate speed,substrate adding time and adding method were optimized.The conversion rate reached 80.5% when the EP concentration was 10 g·L-1,the initial pH value was 5.0,the inoculum amount was 2.0%,the rotate speed was 200 r·min-1,and the adding time was 28 h for EP dissolved in ethanol and treated by ultrasound.In addition,the C11α-hydroxylation of EP in the presence of hydroxypropyl-β-cyclodextrin(HP-β-CD) was also studied,the best molar ratio of HP-β-CD to EP was 1.25∶1,the adding time was 4 h,and the conversion rate reached 93.1%,which increased by 12.6% compared with the control experiment.
