摘 要:目的观察龙血竭对肺纤维化大鼠肺组织转化生长因子β1(TGF-β1)mRNA和I型胶原蛋白表达的影响,探讨其防治肺纤维化的作用机制。方法将30只SD大鼠随机分为模型组、治疗组和正常组各10只。模型组和治疗组大鼠气管内注入博来霉素(5mg/kg)诱导肺纤维化;正常组经气管内注入等容积0.9%氯化钠注射液。从第2d起,治疗组大鼠给予龙血竭180mg/kg灌胃;模型组和正常组以等容积0.9%氯化钠注射液灌胃。所有大鼠于第29d处死。苏木素-伊红(HE)及胶原纤维(VG)染色观察肺组织病理改变和胶原沉积情况;免疫组化链霉菌抗生物素蛋白-过氧化酶连结(SP)法检测I型胶原蛋白的表达;实时荧光定量聚合酶链反应(FQ-CR)检测肺组织TGF-β1mRNA的表达。结果正常组I型胶原蛋白在气管周围弱表达,模型组Ⅰ型胶原蛋白分布于支气管上皮下组织、肺间质及血管周围,且表达较对照组增加(P<0.01);治疗组较模型组Ⅰ型胶原蛋白表达减弱(P<0.01)。模型组TGF-β1mRNA的表达较正常组明显增高(P<0.01),治疗组TGF-β1mRNA的表达较模型组明显降低(P<0.01),而与正常组比较差异无统计学意义(P>0.05)。结论龙血竭可能通过抑制大鼠肺组织TGF-β1mRNA的表达,阻止I型胶原蛋白过度沉积,从而有效地减轻肺纤维化大鼠肺组织的纤维化程度。Objective To investigate the effect of the Dragon's blood on the expression of TGF-β1 mRNA and collagen I in the lung tissue of rats with pulmonary fibrosis,and to evaluate the mechanism of the Dragon's blood on pulmonary fibrosis.Methods 30 SD rats were randomly divided into fibrosis model group,treatment group and normal control group.Pulmonary fibrosis were induced by the intratracheal injection of bleomycin(5 mg/kg) in the model group and the treatment group.In the normal control group,saline was given intratracheally.The Dragon's Blood was administered intragastricly in the treatment group with a dose of 180 mg/kg diluted in 2 mL saline while saline was given intragastricly to the other two groups with the same dozes from day 2 till day 28 after modeling.All rats were sacrificed on the 29 th day.The lung histopathology changes and collagen deposition were observed by HE staining and VG staining.The expression of collagen I was examined by an immunohistochemical staining.Expression of TGF-β1 in the lungs at mRNA levels were detected by real time quantitative FQ-CR.Results The expression of collagen I in the lung tissue of rats in model group were higher compared with the control group(0.96 ±0.38 vs 0.23 ±0.15,P〈0.01) and the expression of collagen I in treatment group was significant lower than that in model group(0.39 ±0.14 vs 0.96 ±0.38,P〈0.01).The expression of pulmonary TGF-β1 mRNA in model group was significant higher compared with the control group(P〈0.01) and the expression of pulmonary TGF-β1 mRNA in treat group was significant lower than that of model group(P〈0.01).Conclusion The Dragon's Blood can effectively improve rats' pulmonary fibrosis through inhibiting the expression of TGF-β1 mRNA and preventing the excessive deposition of collagen I in the lung tissue.
