摘 要:目的探讨Fas/FasL诱导细胞凋亡在大鼠抗肾小球基底膜(GBM)肾炎中的作用。方法复制大鼠抗GBM肾炎模型,在实验室的2、7、14、21和28d,行以下处理:收集24h尿、血清样本检测大鼠24h尿蛋白、血肌酐及血尿素氮含量;取肾组织经光镜、电镜观察肾组织病理变化;采用RT-PCR法和Western blot法检测肾组织中Fas、FasL和Caspase-3的表达情况;采用分光光度法检测肾组织中Caspase-3活性;采用TUNEL法检测大鼠肾组织细胞凋亡情况。结果与正常对照组大鼠相比,肾炎模型组大鼠24h尿蛋白、血肌酐及血尿素氮含量均明显升高(P<0.01);肾组织中Fas、FasL mRNA及其蛋白和Caspase-3 mRNA及其蛋白活性均明显增加(P<0.01);肾组织中凋亡细胞数明显增多(P<0.01),且与Fas、FasL及Caspase-3表达呈正相关(P<0.01)。结论 Fas/FasL诱导的细胞凋亡在大鼠抗GBM肾炎中发挥重要作用。[Objective] To investigate the effect of apoptosis mediated by Fas/FasL on anti-glomerular basement membrane (GBM) nephritis in rats. [Methods] Anti-GBM nephritis model was elaborated by the rabbit anti-rat GBM serum in SD rats. All rats were observed for 24 h urinary protein, serum creatine, serum urea on the 2nd day, the 7th day, the 14th day, the 21st day and the 28th day. At the same time, the kidney specimens were collected at intervals for the observation of the pathological changes. The expressions of Fas, FasL, and caspase-3 in the kidney were detected by RT-PCR and Western blot. Spectmphotometric method was adopted to detect the change of caspase-3 activity during the process of cell apoptosis. Apoptotic cells were determined by TUNEL. [Results] Compared with those in control group, proteinuria, serum creatinine and serum urea nitrogen of rats raised obviously in Anti-GBM nephritis model. The expression of Fas and FasL was significantly increased in the model rats (P 〈 0.05). The expression and the activity of caspase-3 were significantly increased in the model rats (P 〈0.05). Apoptotic index was significantly increased in the model rats (P 〈0.05). Furthermore, a positive correlation existed between nephritic tissue Fas, FasL mRNA and protein, easpase-3 mRNA, caspase-3 activity, the cell counts in the glomeruli and apoptotic index in nephritic rats (P 〈0.05). [ Conclusion ] The apoptosis mediated by Fas/FasL plays an important role in anti-glomerular basement membrane nephritis.
