摘 要:目的探讨阴离子交换蛋白2在人正常绒毛细胞和绒癌JAR细胞中的表达差异,以及该通道蛋白的表达改变与绒癌发生的关系。方法人正常绒毛滋养细胞的原代培养、纯化及鉴定;人绒毛膜癌JAR细胞株的培养。将原代纯化培养的人正常绒毛细胞设为正常组,人绒毛膜癌JAR细胞株设为绒癌组。以免疫组织化学法检测人正常绒毛滋养细胞和绒毛膜癌细胞株JAR细胞膜上AE2的表达。以RT-PCR技术检测人正常绒毛滋养细胞及绒癌JAR细胞中AE2mRNA的表达水平。结果在正常组中,原代培养的人正常绒毛滋养细胞上仅能从显微镜中肉眼观察到AE2的极低表达,以致摄影效果不佳。而在绒癌组中,绒癌JAR细胞膜上AE2的阳性细胞率为(91.59±12.10)%,远高于正常组。在正常人绒毛滋养细胞与人绒癌JAR细胞株中,均检测到AE2mRNA的表达。绒癌组AE2的OD值为(0.85±0.21);正常组AE2的OD值分别为(0.19±0.01)。绒癌组AE2的OD值明显高于正常组(P<0.05)。结论 AE2表达异常上调,滋养细胞内、外的水电解质平衡将被破坏,可能是导致绒癌发生的原因之一。【Objective】 To discuss differential expression of Anion Exchanger 2 in cultured normal human trophocyte and JAR cell line,and the relationship between the changed expression of the channel protein and nosogenesis of chorionic carcinoma.【Methods】①The human cultured normal trophocyte was primary cultured,purified and identified,then it was set as Normal Group.JAR cell line was cultured and setted as Choriocarcinoma Group.② We have found the expression of AE2 in cultured normal human trophocyte and JAR cell line by immunohistochemistry.③ We have found the expression of AE2 in cultured normal human trophocyte and JAR cell line by RT-PCR.【Results】①In Normal group,we could just observe so limited expression of AE2 in cultured normal human trophocyte that we could not take photo of them.But in Choriocarcinoma group.However,the rate of positive cell of AE2 in JAR cell line were(91.95±12.10)%,much higher than that in Normal Group.②we could detect the expression of Anion Exchanger 2 in both cultured normal human trophocyte and JAR cell line.Choriocarcinoma Group AE2,respectively,for the OD value(0.85 ± 0.21),Normal Group,AE2,respectively,for the OD value(0.19 ± 0.01).The OD value of AE2 in Choriocarcinoma Group was significantly higher than that in the Normal Group(P 0.05).【Conclusion】 The overexpression of AE2 may lead to the imbalance of water and electrolyte in trophocyte,and it may be a mechanism of choriocarcinoma.
