杜仲叶和杜仲皮中化学成分的比较

摘　　要：目的:利用杜仲叶代替杜仲皮入药是人们非常关心的问题,探讨它们之间化学成分的差异,及杜仲叶代替杜仲皮入药的可能性.方法:采用系列甲醇-水(50%至100%甲醇含量)为溶剂,对杜仲叶和杜仲皮进行超声提取,然后以正己烷、乙酸乙酯、正丁醇依次进行分别萃取,得到4组(包括水相)不同极性段的化合物.采用C18柱和C8柱进行色谱分析.结果:结果显示,杜仲叶和杜仲皮所含化学成分存在明显的差异,杜仲叶中的化学成分比杜仲皮少许多,而且除少数几个成分外,含量也普遍低于杜仲皮.特别是在正丁醇溶剂中两者组成成分相差明显,叶中至少含有4种皮中不具有的化合物.结论:杜仲叶不含有杜仲皮中全部的化学成分,而且具有杜仲皮不含有的成分,在代替杜仲皮入药时应谨慎.Objective Through investigating the difference in chemical constituents between the leaf and bark of Eucommis ulmoides to explore the possibility of replacing the bark of E.ulmoides with its leaf as a traditional Chinese medicine.Method A series of methanol-water（50%-100% methanol content） was used as solvents to extract E.ulmoides leaf and bark intermittently and repeatedly in an ultrasonic extractor to obtain all the chemical compounds contained in them.Then the samples were extracted,in turn,with n-hexane,acetic ester and n-butyl alcohol,so that chemical compounds differing in polarity were separated accordingly.Chromatographic peaks of chemical compounds associated with different polarities were detected through C18 column and C8 column.Result The comparison of peaks show the compounds resolved in the three solvents and aqueous phase were obviously different between E.ulmoides leaf and E.ulmoides bark.The number of compounds in E.ulmoides leaf was less than that in E.ulmoides bark,and except for a few compounds,the content of most chemical components in E.ulmoides leaf were,generally,lower than in E.ulmoides bark.Especially,obvious difference was detected between the leaf and the bark in their chemical components resolved in n-butyl alcohol.At least four chemical compounds were found to exist in the leaf but not in the bark.Conclusion E.ulmoides leaf does not contain all the chemical components contained in E.ulmoides bark and contains some compounds that E.ulmoides bark does not.Therefore,it must be extremely cautious when we attempt to replace E.ulmoides back with E.ulmoides leaf as medicine.
