摘  要:目的探讨IFN-γ对兔盆腔术中照射后直肠组织的病理变化和转化生长因子-β1(TGF-β1)含量的影响。方法 54只雌性新西兰大白兔随机分成单纯照射组、药物干预组和空白对照组,盆腔术中照射后当天开始每隔4 d给予药物干预组肌注IFN-γ2.5×104u/(kg.f),共5次;于照射后第4、8、12、16、20周每组分别处死5只兔,空白对照组于实验完成时统一处死,光镜下分别观察直肠组织的病理变化,原位杂交法检测TGF-β1mRNA的表达变化。结果光镜下观察直肠组织病理改变及原位杂交TGF-β1mRNA含量,对比药物干预组与单纯照射组和空白对照组间均有显著性差异(P<0.05)。结论 IFN-γ能明显抑制TGF-β1mRNA的表达,可减轻早期放射性炎症反应及后期放射性纤维化的程度。Objective To study the effect of interferon-γ(IFN-γ) on pathological changes and transforming growth factor-β1(TGF-β1) changes in rectal tissues of rabbits which received introoperative radiotherapy(IORT).Methods Fifty-four big female New Zealand albino rabbits were randomly allocated into simple irradiation group,IFN-γ treatment group and blank control group.IFN-γ(2.5×104u/kg·f) was injected into IFN-γ treatment group every four days after IORT for five times.Five rabbits were randomly sacrificed at 4,8 12,16 and 20 weeks after IORT except those in blank control group.Pathological changes of the rectum were observed under light microscope and the level of TGF-β1 mRNA expression was observed by hybridization in situ.Results Light microscopy showed that the pathological changes and the mRNA expression of TGF-β1 in IFN-γ treatment group at different time phases were significantly lower than those in simple irradiation group and blank control group(P〈0.05).Conclusion IFN-γ can significantly inhibit the mRNA expression of TGF-β1,abate acute factitial proctitis and radiation-induced rectal fibrosis.
