摘  要:目的探讨肝性脑病(HE)大鼠肝脏及脑组织中的蛋白质硝基化和细胞凋亡与HE发病的关系。方法雄性Wistar大鼠随机分为HE组和空白对照组。HE组用硫代乙酰胺(TAA)制造大鼠HE模型。采用链霉亲和素-生物素-酶复合物(SABC)免疫组化法检测肝脏和脑组织硝基化蛋白质和一氧化氮合酶(NO S);用原位末端脱氧核糖核酸转移酶标记法(TUNEL)检测细胞凋亡,并计算凋亡指数;空白对照组大鼠用生理盐水做平行对照。结果 HE组大鼠肝脏和脑组织中检测到明显的硝基化蛋白质沉积,而对照组少见;HE组大鼠肝脏和脑组织中检测到NO S的表达,对照组甚少;HE组大鼠肝脏和脑组织细胞凋亡指数较对照组明显增高(P<0.01)。结论 HE与其肝脏和脑组织蛋白质硝基化关系密切。Objective To investigate protein nitrotyrosine and cell apoptosis in the liver and brain tissues of a rat model of hepatic encephalopathy(HE).Methods Male Wistar rats were randomly divided into the HE group and the control group.To produce the HE model,the HE group was treated with thioacetamide(TAA) by intraperitoneal injection.The control group was treated with normal saline in the same way.Then frozen sections were made from the liver and brain of the 2 groups.In the frozen sections,protein nitrotyrosine(3-NT) and nitric-oxide synthase(NOS) was tested by the immunoenzymic method of SABC,and cell apoptosis was tested by TdT-mediated dUTP nick-end labeling(TUNEL).The cell apoptosis index was calculated.Results Significant nitrate protein deposition was observed in the frozen liver sections in the HE group,while it was rare in the control group.Expression of NOS was greater in the liver and brain tissues of the HE group.The cell apoptosis index was significantly higher in the HE group than in the control group(P0.01).Conclusions There is a close relationship between HE and protein nitration in liver and brain tissues.
