改良5'-核苷酸酶-碱性磷酸酶双染色法观察成牙本质细胞层毛细淋巴管

摘　　要：目的:对5'-核苷酸酶-碱性磷酸酶(5'-nucleotidase-alkaline phosphatase,5'-Nase-ALPase)双染色法进行改良,并用其染色观察牙髓成牙本质细胞层是否存在毛细淋巴管。方法:选用牙根发育完成的健康恒前磨牙共44个。首先取15个牙,分为3组,分别用单纯冰冻法、先冰冻后固定法、先固定后冰冻法处理牙髓,选取适用于本研究的标本制备方法。然后取8个牙的牙髓切片对5'-Nase-ALPase双染色法从ALPase反应底物量和反应时间上进行改良。最后取21个牙,其中12例牙髓的切片用改良双染色法染色,9例分3组作对照染色,在光镜下观察成牙本质细胞层中的毛细淋巴管。结果:用改良5'-Nase-ALPase双染色法发现在被染为淡蓝色的成牙本质细胞层中有呈褐色或浅黄色的毛细淋巴管。结论:在牙根发育完成的健康恒前磨牙成牙本质细胞层中有毛细淋巴管。AIM：To modify the 5＇-nucleotidase-alkaline phosphatase（5＇-Nase-ALPase） double staining method and to observe the lymphatic capillaries in the odontoblast layer of pulp with the modified method.METHODS：Healthy premolars with fully developed roots were employed in this study.The 5＇-Nase-ALPase double staining method was modified by reducing the dose of substance and reaction time of ALPase.Twelve premolars were stained with the modified 5＇-Nase-ALPase double staining method and 9 with the control methods to study the lymphatic capillaries in the odontoblast layer.RESULTS：Brown-stained lymphatic capillaries could be observed in the pale-blue odontoblast layer with the modified 5＇-Nase-ALPase double staining method.CONCLUSION：There are lymphatic capillaries in the odontoblast layer of young healthy fully-developed premolars.
