柱后衍生HPLC法测定桑叶中的氨基酸含量

摘　　要：目的建立桑叶中氨基酸的含量测定方法。方法色谱柱:安捷伦zorbax eclipse氨基酸专用柱,柱温:55℃,流速:0.4mL.min-1;衍生试液的流速:0.4mL.min-1,流动相A和流动相B以梯度洗脱;荧光检测波长:激发波长:338nm,发射波长:425nm;进样量:20μL。结果能有效分离出桑叶中的15种氨基酸。结论本法简便,结果可靠,可用于桑叶粗蛋白中的氨基酸含量测定。Objective To establish a HPLC method for the determination of amino acids in folium.Methods The analysis was performed on zorbax eclipse column;the flow rate of the mobile phase was 0.4mL·min-1;the gradient elution was used;the flow rate of the fluorescence reagent was 0.4mL·min-1;the excitation wavelength was 338nm,and the emission wavelength was 425nm.Results 15 amino acids in folium were seperated.Conclusion This method is simple,stable and fast.It can be used for determination of amino acids in folium.
