摘 要:目的为了解健康成人和放射工作者体细胞HPRT基因位点突变率。方法应用人外周血淋巴细胞抗6-TG分析技术检测了30例健康成人和30例放射工作者体细胞HPRT位点突变频率。结果健康成人HPRT基因位点突变频率均值为01628×10-3,变化范围为01300×10-4~02903×10-3,放射工作HPRT基因位点的突变频率均值为02225×10-3,变化范围为0337×10-4~0478×10-3。t检验P<005,两者差异显著。从检测结果亦可看出,随着放射工龄的延长,HPRT基因位点突变频率也随之增加。结论体细胞HPRT基因位点突变频率可作为各种有害因素对机体遗传损伤的生物标识物之一。Objective In order to study mutant frequencies of somatic HPRT locus between healthy adults and radiation workers. Methods Mutant frequencies of somatic HPRT locus in peripheral blood lymphocytes of 30 healthy adults and 30 radiation workers were detected by using 6 thioguanine resistant(TG r)assay. Results The results showed that the TG r mutant frequency in the healthy adults was 0 1628×10 -3 ranged from 0 1300×10 -4 ~0 2903×10 -3 . It was 0 2225×10 -3 ,ranged from 0 337×10 -4 ~0 478×10 -3 in the radiation workers. A significant difference was shown between healthy adults and radiation workers. The TG r mutant frequency also appeared to be increased with the length of radiation service. Conclusion The mutant frequency at HPRT locus can be served as one of the biomarkers for detecting genetic damage of human exposure to environmental and occupational mutagens.
