摘　　要：目的研究ｂｃｌ－２及其表达产物在足叶乙甙诱导的Ｋ５６２细胞凋亡中的作用。方法采用细胞形态观察，ＤＮＡ电泳，ＤＮＡ缺口的原位检测观察Ｋ５６２细胞的凋亡，用原位杂交法检测ｂｃｌ－２ｍＲＮＡ的表达，用免疫荧光法检测ｂｃｌ－２蛋白的表达。结果２０μｇ／ｍｌ的足叶乙甙能诱导Ｋ５６２细胞发生凋亡，在此过程中，ｂｃｌ－２ｍＲＮＡ及ｂｃｌ－２蛋白的表达均下降。结论ｂｃｌ－２及其表达产物的下调在足叶乙甙诱导的Ｋ５６２细胞凋亡中起重要作用。Objective In order to study the role of bcl-2 and its expression product in K 562 cell apoptosis induced by etopside,Methods We observed K 562 cell apoptosis by cell morphologic changes,DNA fragments agarose gel electrophoresis and in situ TDT assay of DNA breaks,We analysed bcl-2 mRNA level with cytohybridyzation in situ technique and detected bcl-2 protein level by immuno-fluorescence.Results Etopside (20 μg/ml)could induce K 562 cell apoptosis.[WT5”HZ]Conclusion In the course of this process,expression of bcl-2 mRNA and bcl-2 protein all reduced.These results suggested that the reduction of bcl-2 gene and its product played an important role in K 562 cell apoptosis.
