摘 要:目的应用超声生物显微镜(UBM)分析高脂饮食喂养的载脂蛋白E基因敲除(Apo E-/-)小鼠的心脏形态和功能,并观察心肌胶原的变化,以探讨胆固醇增高对心脏的影响。方法选择自8周龄起饲以高脂饮食喂养至16周龄和24周龄的Apo E-/-小鼠各6只,以16周和24周龄C57BL/6小鼠各6只为对照组。采用UBM观察并分析小鼠心脏、大血管的结构及血流频谱,对左心收缩及舒张功能进行评价。分析两组小鼠血脂水平,并观察24周龄Apo E-/-小鼠心肌的胶原改变。结果同周龄的Apo E小鼠与对照组比较,UBM显示的心脏、大血管二维径线测值及各瓣口血流脉冲多普勒测值间差异无统计学意义(P>0.05)。Apo E组小鼠血清中总胆固醇(TC)及甘油三酯(TG)水平较同周龄Apo E组小鼠明显升高,差异有统计学意义(P<0.05)。24周龄Apo E-/-小鼠心肌有胶原增加,Ⅰ型胶原增多。结论 UBM可很好地显示小鼠心脏及大血管,高脂饮食Apo E-/-小鼠血脂水平明显升高,24周龄Apo E-/-小鼠心肌有少量胶原,但目前的UBM观测指标难以反映小鼠心肌病理改变。Objective To explore the heart morphology and function of Apo E mice with high cholesterol-fed by ultrasound biomicroscopy (UBM) , and to study the influence of increased cholesterol on heart by observation of myocardial collagen changes. Methods Twelve Apo E mice of 8 weeks age were selected and divided into two groups, then fed to 16 and 24 weeks age with high cholesterol respectively. C57BL/6 mice in 16 and 24 weeks age were selected as control group with 6 in each age group. The structure and flow, systolic and diastolic function of heart were analyzed by UBM, the serum lipid was analyzed in each group, and changes in myocardial collagen were assessed in 24 weeks age Apo E mice. Results Compared with control group, there was no significant difference among all age groups in the measurements of diameter in heart and great vessels and velocity in each valves (P 〉 0.05 ). The total cholesterol(TC) and triglyceride (TG) in high cholesterol group were significant higher than those in control group ( P 〈 0.05 ). Myocardial collagen was increased in 24 weeks age Apo E mice, especially type I collagen. Conclusion UBM can display the heart and great vessels of mice well, the serum lipid is obviously increased in high cholesterol-fed Apo E mice, myocardial collagen is increased in 24 weeks age Apo E mice, but it couldn' t reflect the pathological changes of myocardium in mice by UBM now.
