摘　　要：目的:研究涎腺肿瘤中骨桥蛋白(Osteopontin,OPN) mRNA和蛋白表达水平与临床病理特征之间的关系。方法:应用荧光实时定量RT-PCR和Western blot检测OPN mRNA和蛋白在41例恶性涎腺肿瘤和21例良性涎腺肿瘤及10例正常涎腺组织中表达量,分析OPN mRNA和蛋白表达量与涎腺肿瘤临床病理特征的关系。结果:OPN mRNA和蛋白在恶性肿瘤中的表达量明显高于在良性肿瘤和正常组织中的表达量,差异具有统计学意义(P<0.05)。OPN mRNA表达水平与恶性涎腺肿瘤TNM分期和淋巴结转移密切相关,蛋白表达水平与TNM分期有关(P<0.05),与发病部位、分化程度无明显相关性。结论:恶性涎腺肿瘤中OPN mRNA和蛋白表达量明显增高,并与临床病理特征密切相关。Objective：To investigate the expression of osteopontin （OPN） mRNA and protein and its correlation with clinico-pathological features of salivary gland tumors. Methods：OPN mRNA and protein level were detected by fluorescent quantitative real-time reverse transcription-polymerase chain reaction （RT-PCR）and Western blot in 41 cases of malignant salivary gland tumors,21 of benign salivary gland tumors and 10 of normal salivary gland tissues. The relationship between the relative content of OPN mRNA and protein and clinico-pathological features of salivary gland tumors were analyzed. Results：The differences of OPN mRNA and protein expression levels in various tissues （malignant salivary gland tumors,benign alivary gland tumors and normal salivary gland tissues） were statistically significant,and the mRNA expression levels were correlated significantly with node metastasis and TNMstage,the expression levels of OPN protein were correlated significantly with node metastasis and TNMstage（P0.05）. Conclusion：OPN mRNA and protein were over-expressed in malignant salivary gland tumors,and they were correlated to the clinico-pathological features. Western blot tumors,benign alivary gland tumors and normal salivary gland tissues） were statistically significant,and the mRNA expression levels were correlated significantly with node metastasis and TNMstage,the expression levels of OPN protein were correlated significantly with node metastasis and TNMstage（P0.05）.
