摘 要:目的:通过动物实验探索β-磷酸三钙(β-TCP)复合组织工程骨应用于颌骨缺损修复的可行性。方法:3只杂交犬的双侧下颌骨人工制备3 cm×1 cm×1 cm缺损后,采用自体对照实验方法。实验侧植入骨髓基质细胞和β-磷酸三钙复合物(BMSC/β-TCP),对照侧仅植入β-TCP,在植入后均使用四环素荧光标记。分别于术后1、3、6个月进行CT扫描,之后处死实验犬,制备组织标本,进行HE染色、苦味酸品红染色、四环素荧光检测,并加以评价。对分析数据结果使用SAS 6.0进行统计学分析,P<0.05为有统计学意义。结果:BMSC/β-TCP修复犬颌骨缺损,有大量的编织骨生成;对照组虽然在6个月后也有一定量的骨质形成,但未见到编织骨。四环素标记结果显示,实验组术后6个月,骨质形成速率较对照组高;对照组在最初的3个月,骨质形成明显比实验组低。在第3~6个月期间,实验组和对照组骨质形成均明显加快。结论:BMSC/β-TCP修复犬的颌骨缺损可以形成质量较高的板层骨,而单纯β-磷酸三钙所形成的骨质纤维较紊乱,哈弗氏骨管的数量也较少。Objective: This study was to explore the application of tissue-engineered bone with β-tricalcium phosphate (β-TCP) for the repair of jaw defects through animal model. Methods: A bone defect was made in each side of mandible in 3 canines. The defect on experimental side was implanted with the complex of bone marrow stromal cells and β- tricalcium phosphate (BMSC/β-TCP), and the defect on control side was implanted with only simple β-TCP. 1,3,6 months after implantation, the tetracycline fluorescence technique and CT scanning were done. Tissue sections were prepared for HE, Van Gieson, Masson and tetracycline fluorescence examination. The results were analyzed by SAS 6,0, and the value of P〈0.05 was considered significant. Results: In experiment group, a large number of lamellar bones were formed. There was also a certain amount of bone formation in the control group at 6 months later, but no lamellar bone. Tetracycline marker showed that bone formation rate of the experimental group was higher than that in control group at 6 months later. The control group bone formation was significantly lower than the experimental group at 3-month period. The bone formation process of both experimental and control groups speeded up evidently after 6 months. Conclusion: There were much higher quanlity lamellar hone formation in experiment group than that in control group. The complex of BMSC/β- TCP may be effective to speed up the process of bone healing and produce high-quality of new bone.
