枯草芽孢杆菌BS224的16S rDNA序列测定及系统进化分析

摘  要:利用细菌通用引物,通过PCR的方法,对菌株BS224的16S rDNA基因进行扩增,PCR产物经胶回收试剂盒纯化后测序,测序结果与GenBank上已登录的高同源16S rDNA序列进行比较,并构建系统进化树,结果BS224与Bacillus subtilisstrain ZHA9相似性最高,达到99.38%,与模式菌株Bacillus subtilisstrain 168的相似性也达到98.55%,结合其形态特征、培养特征、生理生化特性的分析结果,可以确定BS224属于枯草芽孢杆菌属。The universal primers of bacteria were used to amplify the 16S rDNA sequence of BS224 by PCR.The PCR products were purified by Agarose Gel DNA Purification Kit and sequenced.The resultant nucleotide sequence was compared with the registered 16S rDNA sequences in the GenBank,then the phylogenetic tree was constructed.BS224 exhibited the highest similarity,99.38% with B.subtilis strain ZHA9,and also displayed 98.55% similarity with the type strain B.subtilis strain 168.According to determination and phylogenetic analysis of 16S rDNA sequene in BS224,as well as its morphological characteristics,cultural characteristics,physiological and biochemical characteristics,we could prove that the strain BS224 belonged to Bacillus subtilis.
