摘  要:目的探讨雷帕霉素对mTOR基因诱导的NI H3T3成纤维细胞增殖的影响。方法以mTOR基因转染小鼠NI H3T3成纤维细胞,以雷帕霉素干预,用MTT法检测细胞增殖情况,用RT-PCR和Western blot方法测定细胞p70S6KmRNA与蛋白的表达。结果 MTT法检测转染mTOR基因后细胞增殖能力增强,雷帕霉素可抑制其增殖;RT-PCR与Western blot检测转染后细胞p70S6K的mRNA与蛋白表达水平明显增加,雷帕霉素可抑制p70S6KmRNA与蛋白表达水平。结论雷帕霉素可能通过抑制mTOR/p70S6K信号通路抑制成纤维细胞增殖。Objective To observe the mechanism and influence of rapamycin on proliferation of NIH3T3 fibroblasts transfected with mTOR gene.Methods NIH3T3 fibroblasts were transfected with mTOR gene and the proliferation of NIH3T3 fibroblasts was examined by MTT assay.mRNA and protein expression levels of p70S6K were examined by means of RT-PCR and Western blot.Results Rapamycin suppressed the proliferation of NIH3T3 cells and the mRNA and protein overexpression levels of p70S6K in NIH3T3 fibroblasts transfected with mTOR gene.Conclusion Rapamycin can suppress the proliferation of fibroblasts and probable by means of inhibiting mTOR/p70S6K signal pathway.
