当归芍药散对拟血管性痴呆小鼠行为学及脑海马神经肽含量的影响

摘　　要：目的:探讨当归芍药散对血管性痴呆小鼠行为学及脑海马降钙素基因相关肽(CGRP)、内皮素1(ET-1)、生长抑素(SS)含量的影响。方法:将二级昆明小鼠随机分为4组:假手术组、模型组、当归芍药散组、脑复康对照组,每组10只,采用脑缺血再灌注的方法复制血管性痴呆小鼠模型,以脑复康为对照,通过水迷宫法分别观测术后14天各组小鼠行为学以及脑海马CGRP、ET-1和SS含量的变化。结果:与假手术组比较,模型小鼠学习与记忆能力下降,表现为游全程时间延长,错误次数增加(P<0.01),脑海马CGRP和SS含量下降,ET-1含量升高(P<0.01);与模型组比较,当归芍药散可使模型小鼠游全程时间缩短,错误次数减少(P<0.01),脑海马CGRP和SS含量升高,ET-1含量下降(P<0.01),优于对照药物(P<0.05)。结论:当归芍药散通过调节脑海马神经肽CGRP、ET-1和SS含量,改善血管性痴呆小鼠学习与记忆能力。Objective：To probe the effect of Danggui Shaoyao San on ethologic and the content of neuropeptide in hippocamp of vascular dementia mouse.Methods：The second order Kunming mice were divided into four groups：sham operation group,model group,Danggui Shaoyao San group,and Naofukang group,ten mice each group,the mouse model of vascular dementia were reproduced through operation of cerebral ischemia reperfusion,and contrast to Naofukang.Then,to detect the praxiological changes of each group mouse by the means of water maze and the content of CGRP,ET-1 and SS in hippocamp on the 14th day after operation.Results：Compared with sham operation group,the ability of vascular dementia model mouse for learning and memory reduced,which showed extending the time for swimming the whole process,and increasing times of mistakes（P〈0.01）;the content of CGRP and SS in hippocamp reduced,the content of ET-1 increased（P〈0.01）.Compared with model group,Danggui Shaoyao San could make the time for swimming the whole process shorten and times of mistakes decreased（P〈0.01）,increase the content of CGRP and SS in hippocamp,and decrease the content of ET-1（P〈0.01）,which were excellent than that of contrast medicine（P〈0.05）.Conclusion：Danggui Shaoyao San could regulate the content of CGRP,ET-1 and SS in hippocamp and improve the learning and memory capability of model mouse.
