摘 要:目的:研究壮骨镇痛胶囊对高转移人乳腺癌MDA-MB-231细胞骨架蛋白p-Ezrin、F-actin和β-tubulin表达水平的影响。方法:RT-PCR法检测不同浓度壮骨镇痛胶囊含药血浆对MDA-MB-231细胞p-Ezrin基因转录水平的影响,免疫细胞化学法检测其对MDA-MB-231细胞p-Ezrin、F-actin和β-tubulin翻译水平的影响。结果:5.0%、1.25%壮骨镇痛胶囊含药血浆显著抑制了MDA-MB-231细胞p-Ezrin基因的转录(P值分别为0.000和0.042)和翻译(P值分别为0.000和0.001),抑制了F-actin(5.0%和1.25%的P值分别为0.003和0.044)和β-tubulin(5.0%和1.25%的P值均为0.000)蛋白表达;0.63%壮骨镇痛胶囊含药血浆反而促进了p-Ezrin基因的转录(P=0.000),但对p-Ezrin、F-actin和β-tubulin蛋白表达无显著影响。结论:壮骨镇痛胶囊可通过抑制乳腺癌MDA-MB-231细胞p-Ezrin、F-actin和β-tubulin蛋白表达来降低细胞的迁移能力,并且该作用与浓度密切相关。OBJECTIVE:To study the effects of Zhuanggu Zhentong capsules (ZZC) on the expression of cytoskeletal proteins such as phos-Ezrin,F-actin and β-tubulin of breast cancer cell lines MDA-MB-231 in vitro.METHODS:The transcriptions of phos-Ezrin in MDA-MB-231 cells treated with different concentrations of ZZC contained plasma were examined by reverse transcriptase PCR protocol (RT-PCR).These cytoskeletal proteins were assayed by immunocytochemistry protocol.RESULTS:The expressions of mRNA (P were 0.000 and 0.042) and proteins of phos-Ezrin (P were 0.000 and 0.001) in the MDA-MB-231 cells treated with 5.0% ZZC contained plasma and 1.25% ZZC contained plasma were inhibited significantly.F-actin and β-tubulin proteins were also inhibited significantly in the cells treated with 5.0% ZZC and 1.25% ZZC contained plasma (F-actin:P were 0.003 and 0.044;β-tubulin:both P were 0.000).The expressions of phos-Ezrin mRNA significantly improved (P=0.000) in MDA-MB-231 cells troated with 0.63% ZZC contrained plasma.But there were no obvious effects on phos-Ezrin,F-actin and β-tubulin proteins in the MDA-MB-231 cells treated with 0.63% ZZC contained plasma.CONCLUSION:ZZC contained plasma can inhibit the migrations of MDA-MB-231 cells in a dose-dependent manner by inhibiting the expressions of cytoskeletal proteins such as phos-Ezrin,F-actin and β-tubulin.
