奥曲肽联合长春新碱对胃癌细胞的抑制作用

摘　　要：目的:探讨生长抑素类似物-奥曲肽(OCT)联合细胞毒化疗药物长春新碱(VCR)对胃癌细胞系SGC-7901的抑制作用和机制。方法:用MTT比色法和免疫组化法分析OCT、VCR和OCT联合VCR对胃癌细胞生长、细胞凋亡调节基因p53表达的影响。结果:当OCT为1×10-5g/L时,对胃癌细胞的抑制作用最明显;OCT联合VCR对胃癌细胞的抑制高于VCR组(P<0.05);OCT上调p53的表达,也可以协同VCR上调p53的表达(vs对照组,P<0.05)。结论:OCT可以通过上调p53的表达诱导胃癌细胞凋亡,联合应用可以增强VCR对胃癌细胞的抑制作用。Objective： To investigate the inhibiting effect and mechanism of somatostatin analogue, octreotide （OCT）, combined with chemotheroputic medicine,vincristine （VCR）, on gastric cancer cell line SGC-7901. Methods： The growth of gastric cancer cells and the expression of apoptosis modulatory gene were evaluated by MTT colorimetric assay and immunohistochemisty. Results：The significantly inhibiting effect was shown on gastric cancer cells when the concentration of OCT reached 1×10^-5g/L as compared with the control group （P〈0.05）.The inhibiting efficiency of OCT combined with VCR was higher than that of VCR used only （P〈0.05）. The expression of p53 was up-regulated by OCT and VCR associated with OCT （P〈0.05, vs control group）. Conclusion： OCT may induce apoptosis by up-regulating the expression of p53 and increase the inhibiting effect of VCR on gastric cancer cells in combined group.
