摘 要:[目的]观察清胆胶囊对胆固醇结石小鼠肝脏过氧化物酶体增殖物激活受体(PPAR)-γ、胆固醇7α-羟化酶(CYP7A1)及核因子(NF)-κB表达的影响。[方法]38只C57BL/6雌性小鼠随机分为正常对照组(n=10)、模型组(n=15)和清胆胶囊组(n=13)。除正常对照组外,模型组和清胆胶囊组小鼠采用高脂饮食诱发法建立胆固醇结石模型。造模过程中,清胆胶囊组小鼠予清胆胶囊0.36 g/(kg.d)灌胃治疗。8周后观察各组小鼠的成石率,并用Western Blotting法检测肝脏中PPAR-γ、CYP7A1及NF-κB的表达。[结果]清胆胶囊组成石率显著降低(P<0.01),小鼠肝脏PPARγ及CYP7A1表达增强,NF-κB表达降低。[结论]清胆胶囊可通过增强肝脏PPARγ及CYP7A1的表达,抑制NF-κB核转位来发挥防治胆固醇结石的作用。[Objective]To observe the effects of Qingdan capsule(QC)on liver perosisome proliferator activated receptor gamma(PPAR-γ),cholesterol 7 alpha-hydroxylase(CYP7A1)and nuclear factor-kappa B(NF-κB)in C57BL/6J mice model with cholesterol gallstone.[Methods]Thirty-eight C57BL/6J mice were randomly divided into normal control group(n=10),model group(n=15)and QC group(n=13).Cholesterol gallstone model was induced by feeding high-cholesterol diet.Mice in the QC group were intragastricly administered QC 0.22g/(kg d).After 8-week treatment,animals were sacrificed and sampled to calculate the incidences of stone formation.The expression of PPAR-γ,CYP7A1 and NF-κB in liver tissues were detected with western blotting.[Results]The incidence rate of stone formation was 73.33% in the model group,0% in normal control group,and 23.08% in the QC group.The incidence rate in the model group was significantly higher than the normal control group and the QC group(P〈0.01).The expressions of PPAR-γ and CYP7A1 were up-regulated while the expression of NF-κB was down-regulated after the treatment of QC.[Conclusion]QC can decrease the incidence rate of stone formation,induce PPAR-γ and CYP7A1,and decrease NF-κB,which may be its important mechanisms in the treatment and prevention of cholesterol gallstone disease.
