摘 要:目的观察高度白酒和红葡萄酒对肝脏的不同作用;分别从能量代谢和氧化应激两个方面阐述高度白酒可引起小鼠的酒精性肝损伤;同时探讨槲芪散治疗酒精性肝损伤的药理学机制。方法将昆明种雄性小鼠随机分为4组,正常组、红葡萄酒组、白酒模型组和槲芪散治疗组。造模14 d后,取血,采用酶标仪比色法测定血清中丙二醛(malondialdehydehyde,MDA)的含量、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的活性;取肝组织,进行HE染色,过碘酸-雪佛氏染色(schiffperiodicacidshiff,PAS染色);酶组织化学染色法检测葡萄糖-6-磷酸酶(glucose-6-phosphatase,G-6-P)、琥珀酸脱氢酶(succinate dehydrogenase,SDH)以及ATP酶(adenosinetriphosphatase,ATPase)的活性。结果白酒模型组MDA含量显著升高(P<0.05),抗氧化酶类SOD和CAT的活性显著减低(P<0.05);能量代谢的相关酶类ATPase、SDH以及G-6-P活性下降;红葡萄酒组HE染色显示红葡萄酒对肝脏无损伤。槲芪散能够显著降低因高度白酒造成的血清MDA含量(P<0.05),提高抗氧化酶SOD(P<0.05)和CAT(P<0.05)的活性;同时能量代谢相关酶的活性提高以及肝糖原储备增加。结论实验表明长期大量酒精摄入能够造成酒精性肝损伤,而红葡萄酒对肝脏无损伤;槲芪散对酒精性肝损伤有明显的干预作用。To observe the different effects of wine spirit and red wine on liver;to interpret the relationship between alcoholic liver disease and hepatoma in terms of oxidative stress and energy metabolism then to explore pharmacology mechanism of Hu Qi San(HQS) intervening the malignant transformation of alcoholic liver disease.KM strain male mice were randomly divided into normal group,white spirit group,red wine group and HQS group.After 14 days,the MDA,SOD and CAT in serum were detected by colorimetry.HE staining,PAS staining,G-6-P staining,SDH staining and ATPase staining were on the liver tissue.In white spirit group,the contents of MDA,the index reflected lipid peroxidation,were significantly increased,the activities of SOD,CAT and the key enzymes about energy metabolism such as G-6-P,SDH and ATPase were restrained.The red wine had no damage on liver.HQS decreased the level of MDA,improved the activities of antioxidant enzymes and the energy metabolism key enzymes;PAS staining revealed an increase of glycogen granules.These experimental results indicated that long-term intake of ethanol causes some damage to liver,but red wine causes no.HQS has intervention effect on alcoholic liver disease.
