摘 要:目的在转录水平上研究外源性神经节苷脂(GM3)、(GD3)对小鼠巨噬细胞抗肿瘤相关因子白细胞介素1β(IL1β)、肿瘤坏死因子α(TNFα)、白细胞介素6(IL6)表达的影响。方法用反转录聚合酶链反应(RTPCR)法和密度扫描半定量PCR产物,比较测定细胞因子IL1β、TNFα、IL6mRNA(信使核糖核酸)表达量。结果单唾液酸神经节苷脂GM3、双唾液酸神经节苷脂GD3对巨噬细胞作用24h后,与未处理组比较,GM3可极大地增加IL1β、IL6、TNFαmRNA的表达,分别在6mg/L、4mg/L和4mg/L时使3种细胞因子增加136、83和464倍。GD3对mRNA的表达仅见微弱调节作用。结论GM3可在转录水平上对上述3种细胞因子进行正调控,且与剂量呈相关性,而GD3则基本上无作用。一定程度上反映了GM3、GD3对巨噬细胞免疫功能的影响。PURPOSE To study the effects of certain ganaliosides on the modulation of the expression of IL 1β、TNFα and IL 6 of mouse macrophage in transcripal level in vitro. METHODS The mRNA transcripts of IL 1、IL 6 and TNF of macrophages stimulated by GM3 and GD3 were detected by semiquantitative reverse transcription polymerase chain reaction(RT PCR). RESULTS Comparing with the control,GM3 could increase the expression of IL 1β、IL 6 and TNFα greatly.The most effective concentrations were 6 mg/L,4 mg/L and 4 mg/L respectively,the increasing folds for these three cytokines were up to 13.6,8.3 and 46.4 However,GD3 either had no effect or only slightly up regulated the expression of these three cytokines. CONCLUSIONS The results metioned above indicate that GM3 can up regulate these three cytokines in transcription level in a dose dependent manner.In contrast,GD3 have no obvious effects on cytokin expression.The alternations of these three cytokines may suggest the influence of the imunological function of mouse marcrophages by GM3 or GD3.
