摘  要:目的通过激光共聚焦扫描显微镜(LSCM)对脑脊液中脑膜癌细胞线粒体进行立体、动态、定量研究,得到脑膜癌线粒体分布特点,为脑膜癌的诊断、鉴别诊断、细胞分化程度和功能状态判断提供依据。方法将标本分为脑膜癌组、阳性对照组、阴性对照组,用荧光探针Mitotrackcr Grccn FM标记线粒体,得到线粒体的断层扫描图像、荧光强度地形立体分布图。结果脑膜癌组线粒体分布与阳性对照组无明显差别,脑膜癌组线粒体分布与阴性对照组有明显差别。结论采用激光扫描共聚焦显微镜技术对脑膜癌的诊断、鉴别诊断、细胞分化程度和功能状态判断有一定的临床意义。Objective To three-dimensionally,dynamically and quantitatively investigate the mitochondria’s distribution of cerebrospinal fluid meningeal carcinoma cells by laser scanning confocal microscope (LSCM). Methods Mitochondria of all cells (meningeal carcinoma cells,positive control group celsl and negative control group cells) were stained with fluorescent probe Mitotracker Green FM,sectional scanograph and fluorescent intensity stercophotography were obtained,and mitochondria were measured individually. Results Compared with positive control group cells,mitochondria’s distribution in cerebrospinal fluid meningeal carcinoma cells showed no difference,while mitochondria,s distribution in cerebrospinal fluid meningeal carcinoma cells was different with that of negative control group cells. Conclusion The experimental works mentioned above may be useful for meningeal carcinoma diagnosis,differential diagnosis,and judgement of cells differentiation level and function.
