摘  要:目的 探讨PKM2与多聚嘧啶串结合蛋白(PTB)在膀胱移行细胞癌中的表达及意义.方法 半定量逆转录-聚合酶链反应(RT-PCR)法测定PKM2和PTB mRNA在不同分级膀胱癌组织和膀胱癌细胞株中表达水平的变化;Western blot法测定PKM1、PKM2和PTB蛋白表达水平的变化;免疫组织化学方法检测膀胱移行细胞癌组织及正常膀胱组织中PKM1、PKM2与PTB的表达差异.结果 PKM2与PTB的mRNA水平和蛋白水平在膀胱移行细胞癌中明显增高,以高分级癌组织和癌细胞株中最为明显.免疫组织化学显示膀胱移行细胞癌组织PKM2阳性率为84.4%(38/45),高于正常膀胱黏膜组织60.0%(27/45),P<0.05;PTB在膀胱移行细胞癌组中的阳性率为95.6%(43/45),高于正常膀胱粘膜组织55.6%(25/45),P<0.05,且PKM2的表达与PTB呈正相关.结论 PKM2与PTB在膀胱移行细胞癌组织中呈高表达,两者的表达呈正相关,提示PTB可能通过对PKM2的选择性剪接而调控膀胱癌细胞的能量代谢.Objective To explore the expressions of tumor M2 pyruvate kinase (PKM2) and polypyrimidine tract binding protein (PTB) and their clinical significance in bladder transitional cell carcinoma (BTCC). Methods The changes of PKM2 and PTB mRNA levels in different grades of BTCC and cell lines were identified by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR), and the changes of PKM1, PKM2 and PTB protein levels were validated by Western blotting. The expressions of PKM1, PKM2 and PTB were also studied by using the SP method of immunohistochemistry in BTCC samples and normal bladder tissues. Results The mRNA levels and the protein levels of PKM2 and PTB increased in the BTCC tissues, obviously in high-grade tumors and cell lines in comparison with those in normal bladder epithelium. The positive rate of PKM2 was 84.4% (38/45) in BTCC tissues, which was significantly higher than that in normal tissues 60.0% (27/45) ( P 〈 0. 05 ). The expression of PTB was higher in the BTCC tissues (95.6%, 43/45) than that in normal tissues ( 55.6%, 25/45, P 〈 0. 05 ).There was a positive correlation between the expressions of PKM2 and PTB. Conclusion PTB may play an important role in the alternative processing of PKM2 to regulate the energy metabolism of bladder cancer cells.
