摘 要:目的探讨组蛋白去乙酰化酶(histone deacetylase,HDAC)1和6蛋白表达检测在卵巢浆液性癌中的临床病理学意义。方法选取147例卵巢病变的存档蜡块标本,其中包括39例卵巢浆液性囊腺瘤26例卵巢浆液性交界性囊腺瘤和82例卵巢浆液性囊腺癌。应用免疫组化EnVision法检测HDAC1和HDAC6基因蛋白在上述卵巢病变组织中的表达水平,并分析其蛋白检测的临床病理学意义。结果 HDAC1和HDAC6蛋白分别为胞核和胞质染色,HDAC1蛋白强阳性表达率在卵巢浆液性囊腺瘤、交界性浆液性肿瘤和浆液性囊腺癌中分别为7.7%(3/39)、65.4%(17/26)和80.5%(66/82);HDAC6蛋白的强阳性表达在39例卵巢浆液性囊腺瘤中完全阴性,而在交界性浆液性肿瘤中为34.6%(9/26)、在浆液性囊腺癌中则高达86.6%(71/86),差异均具有统计学意义。结论 HDAC1和HDAC6蛋白在卵巢癌发生发展过程中起重要作用,对于卵巢浆液性癌的诊断具有重要的辅助意义,有望成为卵巢癌治疗的新靶点。Purpose To investigate the clinicopathological significance of histone deacetylases (HDACs) expression in ovarian serous tumors. Methods Total 147 cases of paraffin embedded ovarian lesion blocks were collected,including 39 cases of serous cystadenoma,26 serous borderline cystadenoma,and 82 serous adenocarcinoma. The protein expression of HDAC1 and HDAC6 was detected by immunohistochemistry,and the clinicopathological significance of HDAC1 and HDAC6 expression in ovarian serous carcinoma was analyzed. Results Immunohistochemical staining showed that the strongly positive rate of HDAC1 protein was 7.7% (3/39),65.4% (17/26),and 80.5% (66/82) in serous cystadenoma,serous borderline cystadenoma,and serous adenocarcinoma,respectively. Similarly,the HDAC6 protein was negative in all 39 cases of serous cystadenoma,however,the strongly positive rate of HDAC6 protein were 34.6% (9/26) and 86.6% (71/86) in serous borderline cystadenoma and serous adenocarcinomas,respectively. The differences were statistically significant. Conclusions HDAC1 and HDAC6 genes play a key role in the progression of ovarian cancer. The detection of HDAC1 and HDAC6 proteins expression can be used as markers for the early diagnosis of ovarian cancer,and they might be the molecular target for the ovarian cancer therapy.
