摘 要:目的探讨基底型乳腺癌中拓扑异构酶Ⅱα(topoisomeraseⅡalpha,TOP2A)基因的扩增或缺失状态。方法采用荧光原位杂交(fluorescence in situ hybridization,FISH)法对53例基底型乳腺癌和对照组33例管腔A型乳腺癌石蜡包埋标本进行TOP2A基因检测。结果 53例基底型乳腺癌中,TOP2A基因异常者21例,其中扩增14例(4例为簇状扩增、10例为点状扩增)、缺失7例;TOP2A基因正常者32例。管腔A型乳腺癌TOP2A基因1例扩增、6例缺失。基底型乳腺癌中TOP2A基因异常的发生率高于管腔A型乳腺癌(P<0.05)。结论基底型乳腺癌中存在较高比例的TOP2A基因异常,可作为蒽环类化疗药物的治疗靶点。Purpose To investigate the amplification or deletion status of topoisomeras2A(TOP2A)gene in basal type breast cancer.Methods The amplification or deletion status of topoisomeras2A gene were detected in 53 cases of basal type breast cancer and control group 33 cases of luminal A type breast cancer by using fluorescence in situ hybridization(FISH).Results Among the 53 cases,14 cases were showed amplification(4 cases showed cluster pattern of gene amplification and 10 cases showed punctate pattern of gene amplification).7 cases were showed gene deletion.32 cases were showed no TOP2A gene abnormality.In control group,1 case were showed TOP2A gene amplification and 6 cases were showed TOP2A gene deletion.The alterations of TOP2A gene of basal type breast cancer was higher than luminal A type breast cancer(P0.05).Conclusions There were a high rate alterations(amplification and deletion)of TOP2A gene in basal type breast cancer.To detect the TOP2A gene status may help to provide a foundation for choosing anthracycline-containing chemotherapy in basal type breast cancer.
