摘  要:目的探讨大鼠角膜碱烧伤后基质细胞衍生因子-1(stromal cell-derived factor-1,SDF-1)及其CXC受体4(CXC chemokine receptor 4,CXCR4)在角膜组织中的表达变化规律,以及CXCR4特异性拮抗剂AMD3100对角膜新生血管的影响。方法采用1mol·L-1氢氧化钠溶液建立SD大鼠角膜碱烧伤模型,用RT-PCR和Western blot检测大鼠角膜碱烧伤后不同时间点SDF-1和CXCR4的mRNA和蛋白的表达,用免疫组织化学SP法检测大鼠角膜碱烧伤后SDF-1和CXCR4在角膜组织中的阳性表达。另设碱烧伤对照组和碱烧伤治疗组,碱烧伤后分别给予0.1mL生理盐水和AMD3100球结膜下注射,比较2组大鼠角膜新生血管面积和角膜组织中血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白的含量。结果碱烧伤后SDF-1阳性表达于浅层角膜基质细胞、角膜上皮细胞和炎性浸润细胞,CXCR4阳性表达于炎性浸润细胞、角膜上皮细胞和血管内皮细胞。正常大鼠角膜组织中可检出微量SDF-1和CXCR4 mRNA和蛋白表达;碱烧伤后2d,SDF-1 mRNA和蛋白水平明显升高,7d、10d分别是正常水平的3.8倍和21.0倍;CXCR4 mRNA和蛋白水平在碱烧伤后2d明显升高,7d、10d分别是正常水平的1.9倍和17.5倍。碱烧伤治疗组碱烧伤后7d、10d、14d时新生血管面积均小于碱烧伤对照组,差异均有统计学意义(均为P<0.05)。碱烧伤治疗组碱烧伤后2d、5d、7d、10d、14d角膜组织中VEGF蛋白的含量均稍低于对照组,差异均无统计学意义(均为P>0.05)。结论 SDF-1与其受体CXCR4的相互作用可能参与了碱烧伤诱导的角膜新生血管形成,AMD3100能有效抑制角膜新生血管的形成,有可能成为治疗角膜新生血管的方法之一。Objective To investigate the expression changes of stromal cell-derived factor-1(SDF-1) and CXC chemokine receptor 4(CXCR4) in rat corneas and effect of CXCR4 specific antagonist AMD3100 on corneal neovascularization.Methods Corneal alkali burn models of SD rats were established with 1 mol·L-1 sodium hydroxide.RT-PCR and Western blot method were used to detect expression of SDF-1 and CXCR4 mRNA and protein at different time points after alkali burn in rat retinas.Immunohistochemical staining was used to detect distribution of SDF-1 and CXCR4 protein in various layers of retinas after alkali burn.Then alkali burn control group and treatment group were set up,and treated with sub-conjunctival injection of 0.1 mL normal sodium and AMD3100.Retinal neovascularization area and concentrations of retinal vascular endothelial growth factor(VEGF) were compared between the two groups.Results After injury,expression of SDF-1 and CXCR4 were found in inflammatory infiltration cells,corneal epithelial cells and neovascular endothelial cells in retinas.Seldom SDF-1 and CXCR4 mRNA and protein were found in retinas of normal rats.SDF-1 mRNA and protein levels were obviously increased at 2 days after alkali burn,and were 3.8 times and 21.0 times of basic level at 7 days and 10 days after alkali burn.CXCR4 mRNA and protein levels were significantly increased at 2 days after alkali burn,were 1.9 times and 17.5 times of basic level at 7 days and 10 days after alkali burn.In treatment group,neovascularization area was obviously lower than that of control group at 7 days,10 days and 14 days after alkali burn,there were statistical differences(all P0.05);The level of VEGF protein was lower than that of control group at 2 days,5 days,7 days,10 days and 14 days,no difference was found(all P0.05).Conclusion The interaction of SDF-1 and CXCR4 secreted from inflammatory cells may play a role in corneal neovascularization induced by alkali burn.AMD3100 can effectively inhibit corneal neovascularization and can be a way for
