摘 要:将大鼠以高脂喂养结合小剂量链脲佐菌素(STZ)诱导,取血测皮质酮和促肾上腺皮质激素(ACTH)的节律后,留取下丘脑和垂体,用实时定量PCR观察下丘脑和垂体糖皮质激素受体和1113-羟类固醇脱氢酶1(11β-HSD1)mRNA表达的改变。对照组、单纯肥胖组和肥胖糖尿病组大鼠的ACTH和皮质酮的水平没有明显改变(P=0.07),但肥胖组和肥胖糖尿病组皮质酮的节律消失。下丘脑糖皮质激素受体mRNA的表达组间无差异,但肥胖糖尿病组11β—HSD1 mRNA的表达高于对照组(P〈0.05)。垂体糖皮质激素受体和11β—HSD1 mRNA的表达肥胖糖尿病组低于肥胖组,肥胖组又低于对照组(均P〈0.05)。上述结果提示肥胖伴糖尿病大鼠的负反馈调节机制受损可能与垂体11β—HSD1和糖皮质激素受体的表达下降有关。Diabetic rat model was induced by high fat diet combined with streptozotoein (STZ). After the model was established, blood samples were taken from jugular veins to examine plasma adrenoeorticotropic hormone (ACTH) and corticosterone, and hypothalamus and pituitary were removed for real-time PCR. There were no significant differences in basal plasma ACTH and eorticosterone level among control, obese, and obese diabetic rats (P = 0.07 ). The corticosterone rhythm in obese and obese diabetic rats was impaired. Hypothalamus glueoeorticoid receptors (GR) mRNA expressions yielded similar results in the groups, but 11β-HSD1 mRNA expression in obese diabetic rats was up-regulated (vs control rats, P〈0.05). The expressions of GR and l 1β-HSD1 in pituitary of obese diabetic and obese rats were significantly down-regulated (both P〈0.05). In the obese diabetic rats, the impaired glueocorticoid negative feedback was partly due to down-regulation of 11 β-HSD1 and GR expressions in pituitary.
