摘  要:目的:观察转化生长因子(TGF)-β1对绒毛膜癌JEG-3细胞增殖能力及其Smad3、Smad7蛋白表达的影响。方法:体外培养人绒毛膜癌JEG-3细胞,分别给予不同剂量及不同作用时间的TGF-β1进行诱导,四甲基氮唑比色法检测细胞增殖能力变化,蛋白印迹法检测Smad3、Smad7蛋白的表达变化。结果:JEG-3细胞对TGF-β1的诱导作用表现为良好的浓度效应和时间效应。100、200μg/LTGF-β1作用后,细胞增殖率显著增加(P<0.01),Smad3蛋白的相对表达量增加(P<0.05或P<0.01),Smad7蛋白无明显改变(P>0.05);同时100μg/LTGF-β1作用12h后,细胞增殖率显著增加,Smad3蛋白相对表达量增加(P<0.05或P<0.01),Smad7蛋白表达无改变(P>0.05)。结论:外源性TGF-β1能够促进绒毛膜癌JEG-3细胞的增殖,具有良好的浓度效应和时间效应;绒毛膜癌JEG-3细胞逃逸TGF-β1介导的生长抑制作用,可能是由于TGF-β1/Smads信号通路中的Smad3、Smad7蛋白发生异常而致。Objective: To investigate the effect of transforming growth factor (TGF) -β1 on the proliferation and expression of Smad3, Smad7 protein in JEG-3 cells. Methods: The human choriocarcinoma cell line JEG-3 cultured in vitro was given different concentrations and different action times of TGF-β1. The proliferation of JEG-3 cells was measured by MTT assay. The expression of Smad3, Smad7 proteins was measured by Western blotting. Results: JEG-3 cells showed the excellent concentration and time effect responded to the external TGF-β1. The JEG-3 cell proliferation was enhanced after TGF-β1 induction with 100 and 200 μg/L respectively (P 0.01), meanwhile, The expression of Smad3 protein was increased significantly(P 0.05 or P 0.01), but the expression of Smad7 protein showed no change(P 0.05). At the same time, JEG-3 cell proliferation was significantly increased after 12 h 100 μg/L TGF-β1 induction (P 0.05 or P 0.01). The expression of Smad3 protein was increased significantly(P 0.05 or P 0.01), but Smad7 protein expression showed no difference(P 0.05). Conclusion: The external TGF-β1 can stimulate the proliferation of JEG-3 cells, and showing good concentration and time effect. Smad7 protein showed lack of expression and Smad7 protein was expressed at abnormally high levels, which suggested that JEG-3 cells escaped TGF -β1 -mediated growth inhibition may be due to Smad3, Smad7 protein of TGF -β1/Smads pathway.
