摘 要:目的建立大鼠溃疡性结肠炎模型,观察甘草酸二铵(DG)对大鼠溃疡性结肠炎(UC)的疗效及其对肠组织Fas/FasL蛋白表达的影响,继而从细胞凋亡方面来探讨甘草酸二铵治疗大鼠溃疡性结肠炎的作用机制。方法用2,4,6-三硝基苯磺酸(TNBS)灌肠复制大鼠UC模型。健康雌性SD大鼠分为正常对照组、模型组、甘草酸二铵组,每组10只。观察疾病活动指数(DAI)、组织学变化,应用免疫组化(IHC)检测大鼠肠组织Fas/FasL蛋白的表达。结果正常对照组、模型组和甘草酸二铵组的DAI评分分别为0.3±0.8,7.2±0.1,3.2±0.7;组织学损伤评分分别为0.9±0.4,6.8±0.7,3.3±0.5;Fas阳性率分别为6.81±2.20,16.62±2.20,10.23±3.20;FasL阳性率分别为6.02±3.20,17.11±3.10,11.02士3.10;与模型组相比,甘草酸二铵组Fas/FasL的表达显著降低,经one—way ANOVA及SNK—q检验,差异有统计学意义(P〈0.05)。结论甘草酸二铵对TNBS/乙醇诱导的大鼠溃疡性结肠炎治疗作用良好,通过下调Fas/FasL的表达减少上皮细胞的凋亡可能是其机制之一。Objective To develop a model of ulcerative colitis (UC ) in rats and evaluate the role of diammonium glycyrrhizinate ( DG) in experimental colitis and to investigate the function and mechanism of DG in rats with ulcerative colitis. Methods Colitis in DG group and model group was induced by administrating TNBS rectally. Spragur-Dawley( SD) rats were randomized divided into normal control group,model group and DG group(10 rats in each group). Disease activity index( DAI) and histological damage were observed. The expression of Fas/FasL in colon tissues was detected by immunohistochemistry. Results In normal control group, model group and DG group, the DAI were 0.3 ± 0.8,7.2 ± 0.1 and 3.2 - 0.7 respectively ; the histological damage scores were 0.9 ± 0.4,6.8 ± 0.7 and 3.3 ± 0.5 respectively ; the positive percentages of Fas were 6.81 ± 2.20,16.62 ± 2.20,10.23 ± 3.20 respectively ; the positive percentages of FasL were 6.02 ± 3.20,17.11 ± 3.10,11. 02 ± 3. 10 respectively. Compared with model group, the DAI and histological damage scores of colon tissues in DG group were significantly improved ( P 〈 0.05 ) , and the expression of Fas/FasL in DG group decreased significantly ( P 〈 0.05 ). Conclusion The present study indicates that DG can relieve colitis in rats induced by TNBS. Decreasing the expression of Fas/FasL in colon tissues may be the possible mechanism.
