摘 要:目的研究外源性雌激素诱导雄性肥胖大鼠青春期前瘦素水平及相关指标对肥胖的影响及作用方式。方法6周龄雄性SD大鼠每日肌肉注射苯甲酸雌二醇(E2B),持续作用2周,建立肥胖动物模型。于末次注射5d后测量血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL—C)、低密度脂蛋白胆固醇(LDL—C)、甘油三酯(TG)浓度及血清瘦素、雌二醇、睾酮水平,观察各指标对肥胖的影响大小及作用方式。结果实验组肥胖大鼠血清TC,LDL-C以及TG浓度、血清瘦素、雌二醇水平均较对照组大鼠明显增高,差异有统计学意义(P〈0.01),血清HDL—C浓度及睾酮水平较对照组大鼠有所减低,差异有统计学意义(P〈0.01);通径分析结果显示瘦素抵抗导致大鼠肥胖的通径系数为0.1926(P〈0.01);脂肪代谢紊乱是研究因素中导致大鼠肥胖的最主要的因素,其中,总胆固醇、甘油三酯及低密度脂蛋白导致大鼠肥胖的通径系数分别为0.2025(P〈0.01),0.4477(P〈0.01),0.7038(P〈0.01);性激素代谢失衡导致肥胖的通径系数为0.1463(P〈0.01)。结论瘦素抵抗、脂肪代谢紊乱及性激素代谢失衡是导致外源性雌激素诱导青春期前雄性大鼠肥胖的主要因素,确定系数R2=0.6323,并且主要通过直接作用,其中脂肪代谢紊乱对肥胖的影响更为显著。Objective To explore the effect and style of leptin and other relative factors in obesity in prepuberty male rats by morphometry. Methods Male rats aged 6 weeks were treated with estradiol benzoate( E2 B ) by intramuscular injections for two weeks to establish the animal model of obesity. At the fifth clay after the last injection the serum concentration of total cholesterol(TC) , high density lipoprotein-C ( HDL-C ), low density lipoprotein-C ( LDL-C ), triglycerides ( TG ), estradiol, leptin and testosterone were detected. The effect and the style of these factors in obesity were analysed. Result The serum concentration of TC, LDL-C, TG, the serum of blood leptin and estradiol in the experimental group were higher than that in control group, but the serum concentration of HDL-C and testosterone in the experimental group were lower than that in control group( P 〈0.01 ). Path analysis showed that the path coefficient of leptin resistance in obesity in rats was 0. 1926 ( P 〈 0.01 ) ; Fat metabolism is the most important factor leading to obesity, including TC ( path coefficient 0. 2025, P 〈 0.01 ) , blood TG( path coefficient 0.4477 ,P 〈0.01) and LDL( Path coefficient 0. 7038 ,P 〈 0.01 ) ;The path coefficient of metabolic imbalance of sex hormones in obesity in rats was 0. 1463 ( P 〈 0.01 ). Conclusions Our findings demonstrate leptin resistance, fat metabolism disorder and metabolic imbalance of sex hormones are the main factors for xenoestroge-induced obesity in prepuberty rats, the coefficient of determination R2 = 0. 6323. Moreover, fat metabolism disorder is the more important one in these factors.
