摘 要:目的:观察嗅鞘细胞条件培养液(OECCM)和星形胶质细胞条件培养液(ACM)对受损海马神经元的保护作用。方法:分别采用2种胶质细胞条件培养液预处理受损海马神经元(谷氨酸诱导损伤),对比其对受损海马神经元活力、凋亡率和胞浆内游离钙离子浓度的影响。结果:与谷氨酸组相比,OECCM组神经元活力明显增高(P<0.01),ACM组神经元活力增高(P<0.05);与OECCM组相比,ACM组神经元活力降低(P<0.05)。与谷氨酸组相比,ACM组神经元凋亡率和胞内游离钙浓度降低(P<0.05),OECCM组明显降低(P<0.01);OECCM组神经元凋亡率和胞内游离钙浓度低于ACM组(P<0.05)。结论:OECCM和ACM均可明显提高受损海马神经元活力、降低凋亡率和胞浆内游离钙离子浓度,OECCM的保护作用优于ACM。Objective: To evaluate the neuroprotection effects of olfactory ensheathing cells conditioned medium(OECCM) and astrocytes conditioned medium(ACM) on injured hippocampal neurons.Methods: Hippocampal neurons which were individually pretreated by ACM or OECCM have been injured by glutamate.Neuronal viability,apoptosis rate and intracellular free calcium concentration have been measured.Results: Compared with that in the glutamate group,the neuronal viability of neurons pretreated either by OECCM or by ACM,was increased and the apoptosis rate and intracellular free calcium concentration were reduced.However,the neuron viability in OECCM group was higher than that in ACM group and the apoptosis rate and intracellular free calcium concentration were lower than that in ACM group.Conclusion: OECCM and ACM pretreatment could remarkably increase the viability and reduce the apoptosis rate and the intracellular free calcium concentration of injured hippocampal neurons and OECCM could provide a more promising condition for neurons.
