摘 要:目的探讨肾细胞癌(RCC)组织及癌旁组织中E-钙黏附蛋白及E-钙粘连素(CDH1)基因的表达与RCC发病机制间的相关性。方法 RCC患者36例(RCC组),男24例,女12例;分别取血液和肾癌组织标本。对照组36例为非肿瘤患者,男30例,女6例,取血液标本。癌旁组织标本为对照组,ELISA方法测定血浆中CDH1定量,免疫组化方法检测肾癌组织及癌旁组织CDH1的表达情况。竞争性PCR方法半定量分析组织中CDH1表达。分析RCC临床特点与CDH1表达的关系。结果 RCC组血浆中CDH1(4.3±1.8)ng/ml较对照组(6.8±3.1)ng/ml低(P<0.05)。免疫组化显示肾癌组织中CDH1表达阳性率为36.1%(13/36),癌旁组织中的表达阳性率为69.4%(25/36),肾癌组织与癌旁组织中CDH1表达比较差异均有统计学意义(P<0.05)。肾癌组织CDH1总RNA和mRNA水平与癌旁组织比较差异有统计学意义(P<0.05)。结论 CDH1基因表达下调可能在肾透明细胞癌的发生中发挥重要作用。Objective To investigate the correlation between the expression of E-cadherin and E-cadherin ( CDH1) gene in the renal cell carcinoma ( RCC) as well as adjacent tissues and the pathogenesis of renal cell carcinoma. Methods The blood specimens and kidney tissue of 36 patients with RCC,including 24 males and 12 females,were collected,and the blood specimens in 36 patients without tumor,including 30 males and 6 females were collected. The adjacent tissue of renal cell carcinoma was used as control group. The ELISA method was used to detect quantitatively the plasma levels of CDH1,and immunohistochemical method was used to determined the expression of CDH1 in RCC and adjacent tissues. The expression of CDH1 in tissues was detected by competitive RT-PCR. The data of 2 groups were compared and the correlation between clinical characteristics of RCC and the expression of CDH1 was analyzed. Results The expression of CDH1( 4. 3 ± 1. 8) ng/ml in patients with RCC was significantly lower than that ( 6. 8 ± 3. 1) ng/ml of patients without tumor ( P 0. 05) . The positive rate of CDH1 expression in RCC patients was 36. 1% ( 13 /36) ,which in cancer adjacent tissue was 69. 4% ( 25 /36) ,there was a significant difference in the expression of CDH1 between RCC group and control group ( P 0. 05) . There was a significant difference in total RNA and mRNA levels of CDH1 between RCC tissues and cancer adjacent tissues ( P 0. 05) . Conclusion The downregulation of CDH1 gene expression may play an important role in the incidence of RCC.
