摘 要:目的观察华蟾素注射液联合化疗小鼠原位移植性肝癌的抑制作用。方法将40只BALB/C小鼠随机分为模型组,单纯化疗组(5-氟尿嘧啶,5-Fu),华蟾素注射液组,化疗(5-Fu)+华蟾素注射液组,每组10只。进行原位移植H22肝癌造模。给药结束第3天,摘取小鼠肿瘤组织和胸腺组织计算抑瘤率和胸腺指数,用流式细胞仪观察肿瘤细胞周期及细胞凋亡情况,取血测定血清肿瘤坏死因子-α(TNF-α)。结果华蟾素注射液组和单纯化疗组均可以明显抑制肿瘤生长,化疗+华蟾素注射液组使抑瘤作用增强;华蟾素注射液组可以增加小鼠胸腺重量及胸腺指数(P<0.01);与模型组比较,单纯化疗组的肿瘤细胞G2/M期细胞比例增加(P<0.05),华蟾素注射液组的肿瘤细胞S期细胞比例增高(P<0.05),化疗+华蟾素组的肿瘤细胞G2/M期、S期细胞比例均高于模型组。结论 5-Fu联合华蟾素可以增加对小鼠原位移植H22肝癌的抑瘤作用,并能减轻用5-Fu进行化疗时产生的毒性反应。Objective To observe the therapeutic effect of Huachansu combined with chemotherapy on mice with orthotopic transplantation hepatocellular carcinoma ( H22 ) . Methods Forty BALB/C mice were randomly divided into control group,fluorouracil group,Huachansu( HCS) group and fluorouracil plus HCS group. After the models of orthotopic transplantation hepatocellular carcinoma ( H22) were established in the mice. The normal saline( 0. 2ml) ,fluorouracil ( 30 mg/kg) ,HCS u( 5ml/kg) ,fluorouracil ( 30mg/kg) plus HCS ( 5ml/kg) were injected intraperitoneally,respectively in the mice of four groups. groups. After the drugs had been administered completely for there days,all the mice were sacrificed,the tumors and the thymus were weighed. The inhibition tumor rates and thymus index were calculated and compared among the four groups. The cell cycle and cell apoptosis were detected by flow cytometry. The content of TNF-α in serum was examined by desmoglein enzyme linked immunosorbent assay ( ELISA) . Results The inhibition effect of tumor growth in fluorouracil group and HCS group was obvious. The inhibition effect was more obvious in fluorouracil plus HCS group. HCS was able to increase the weight of thymus and the thymus index. As compared with control group,the percentage of S phase of cell cycle was increased in HCS group and the percentage of G2 /M phase was increased in fluorouracil group. Both of the percentages were increased in fluorouracil plus HCS group. Conclusion There exists synergistic inhibitory effect between HCS and fluorouracil on hepatocellular carcinoma. The toxicity of fluorouracil is decreased by HCS.
