摘 要:目的观察肉桂酸(CINN)和1,25-二羟维生素D3(1,25-(OH)2D3)联合用药对人胃腺癌MGC-803细胞的诱导分化作用。方法 1,25-(OH)2D3和CINN联合用药(2 mmol/L CINN+10-6mmol/L 1,25-(OH)2D3、1 mmol/L CINN+10-5mmol/L 1,25-(OH)2D3)及单独用药作用于MGC-803细胞48 h后,应用四甲基偶氮唑蓝(MTT)法测定细胞增殖能力,平板克隆试验测定细胞集落形成率,流式细胞仪测定细胞周期,TRAP-银染法测定端粒酶活性。结果 1,25-(OH)2D3和CINN联合用药后胃腺癌细胞生长明显受抑制,细胞周期出现向G0/G1期移行的特征性动力学改变,端粒酶活性明显受到抑制。细胞集落形成率明显下降。结论 CINN和1,25-(OH)2D3联合用药对人胃腺癌细胞的诱导分化具有协同作用。Objective It is to observe the effects of 1,25-(OH)2D3 combined with cinnamic acid(CINN) on differentiation of MGC-803 cells.Methods MGC-803 cells were treated by 1,25-(OH)2D3 and cinnamic acid(2 mmol/L CINN+10-6 mmol/L 1,25-(OH)2D3,1 mmol/L CINN+10-5 mmol/L 1,25-(OH)2D3 and simple use respectively) for 48h,and the rate of growth suppression was evaluated by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay.Plate clone formation assay was carried out to detect the phenotypes of colony formation.FACS analysis was used to analyze the cell cycle.The activity of telomerase was examined by telomeric repeat amplification protocol(TRAP) PCR-silver staining.Results 1,25-(OH)2D3 and cinnamic acid showed significant inhibitory effect on growth of these gastric carcinoma cells at 48 h.After treated with 1,25-(OH)2D3 and cinnamic acid for 48 h,cell cycle showed G0/G1 phase arrest,and the activity of telomerase was inhibited significantly.The colony-forming rate was reduced drastically.Conclusion: 1,25-(OH)2D3 combined with cinnamic acid can induce differentiation of MGC-803 cells synergistically.
