摘 要:目的:建立银屑灵流膏(当归、赤芍、川芎等)中绿原酸、芍药苷、异秦皮啶、阿魏酸和落新妇苷5种成分含量的测定方法,控制产品质量。方法:采用HPLC-PDA法,色谱柱为Kromasil100-5C18,流动相为甲醇-0.1%冰醋酸,梯度洗脱,梯度为0~10min(10∶90~30∶70),10~40min(30∶70~40∶60);流速为1.0mL/min;检测波长绿原酸、异秦皮啶、阿魏酸为320nm,落新妇苷为289nm,芍药苷为230nm。结果:上述条件下5种成分分离良好,绿原酸在2.66~31.86μg/mL(r=0.9999),芍药苷在34.95~419.37μg/mL(r=0.9999);异秦皮啶在0.96~11.55μg/mL(r=0.9999);阿魏酸在0.82~9.84μg/mL(r=0.9999);落新妇苷在6.58~78.92μg/mL(r=0.9999)范围内线性关系良好,5种成分的加样回收率在96.6%~103.7%之间;RSD小于3.83%。结论:该方法方便、准确、灵敏度高,可作为银屑灵流膏中绿原酸、芍药苷、异秦皮啶、阿魏酸和落新妇苷5种成分的质量控制方法。AIM: To establish a quantification method for determining chlorogenic acid,peoniflorin,isofraxidin,ferulaic acid and astilbin in Yinxieling liquid extract(Radix Angelicae sinensis,Radix Paeoniae rubra,Rhizoma chuanxiong,etc. ),and to control the quality of the preparation. METHODS: High performance liquid chro-matography-photodiode array (HPLC-PDA) method was developed in this study. The column was Kromasil 100-5 C18,the mobile phase consisted of methanol-0. 1% acetic acid(10 ∶ 90-30 ∶ 70) during 0-10 min,and (30 ∶ 70 ~ 40 ∶ 60)during 10-40 min in gradient elution with flow rate at 1. 0 mL/min. The detection wavelength was set at 320 nm for chlorogenic acid,isofraxidin and ferulaic acid,289 nm for astilbin and 230 nm for peoniflorin. RESULTS: The calibration curves of the five components were excellent,the linear ranges of chlorogenic acid,peoniflorin,isofraxidin,ferulaic acid and astilbin were in the ranges of 2. 66-31. 86 μg/mL(r = 0. 999 9),34. 9-419. 37 μg/mL(r =0.999 9),0.96-11.55 μg/mL(r =0.999 9),0.82-9.84 μg/mL(r =0.999 9),6.58-78.92 μg/mL (r =0.999 9),respectively. The average recoveries ranged from 96.6% to 103.7%,the relative standard deviations were less than 3. 83% . CONCLUSION: The established method is convenient and accurate with a satisfactory separation and high sensitivity. It is suitable for the quality control of the five components in Yinxieling liquid extract.
