摘 要:[目的]为鞘氨醇单胞菌属的开发利用提供依据。[方法]通过菌株培养、BIOLOG微生物鉴定系统鉴定和16S rRNA序列同源性分析,对1株具有农药降解潜能的鞘氨醇单胞菌XJ进行鉴定。[结果]XJ菌株在LB和MAC培养基上的生长、革兰氏染色反应、氧化酶和TSI试验结果表明,该菌株为革兰氏阴性非肠道菌;对BIOLOG微生物鉴定系统中的95种供试碳源,XJ不能利用的碳源有37种(包括多羟醇和菊糖),符合鞘氨醇单胞菌的生理生化鉴别特性;XJ菌株可扩增出1500bp左右的16S rRNA基因片段,且其16S rRNA基因序列与登陆号为AF331661(Sphingomonas yanoikuyae)的菌株最接近。[结论]供试菌株被鉴定为矢野口鞘氨醇单胞菌XJ菌株(S.yanoikuyaeXJ)。[Objective] The purpose of the study was to provide basis for exploitation of Sphingomonas yanoikuyae.[Method] Through strain culture,identification by using BIOLOG microbiological identification system and 16S rRNA sequence homological analysis,a Sphingomonas yanoikuyae strain XJ with potency to degrade pesticides was identified.[Result] The results from experiments on growth of the strain XJ on media LB and MAC,gram staining reaction,oxidase and TSI tests showed that this strain was gram-negative non-intestinal bacterium;37(including polyhydroxyl alcohol and inulin)of 95 tested carbon sources in BIOLOG microbiological identification system were unavailable for XJ,this was consistent with physiological and biochemical identification characteristics of S.yanoikuyae;16S rRNA gene fragment with length of about 1 500 bp could be amplified from the strain XJ and its 16S rRNA gene sequence was closest to that of S.yanoikuyae strain with logging number of AF331661.[Conclusion] The tested strain was identified as S.yanoikuyae XJ.
