摘  要:[目的]分析集胞藻6803细胞培养体系的光衰减及分批培养条件下藻细胞的生长特性。[方法]以集胞藻6803(Synechocystis sp.PCC6803)为供试藻种,用尤尼柯7200型分光光度计测定藻细胞浓度和用ST-85自动量程照度计测定光强在通过不同光程、不同浓度藻液时的变化。[结果]随着藻细胞浓度和光程的增加,光强迅速下降。在同一光程下,光强随着藻细胞浓度的增加而下降。在培养初期藻细胞浓度较低时,光衰减程度较小;在培养后期,随着藻细胞浓度的增加,光衰减程度逐渐增加。在培养过程中藻体细胞的生长规律与微生物发酵过程中菌体的生长规律相似。藻体细胞生物量曲线与微生物发酵时的菌体生长曲线相似。[结论]Lambert-Beer公式Ln(I/I0)=-KaXL能较好描述藻细胞浓度和光程对光衰减的综合影响;Logistic方程能很好描述藻细胞的生长曲线。[Objective] The purpose was to analyze the light attenuation of cell culture system of Synechocystis sp.PCC 6803 and the growth characteristics of alga cell under batch culture condition.[Method] With Synechocystis sp.PCC 6803 as tested alga strains,the alga cell concn.was determined by using spectrophotometer Type Unico7200 and the changes of light intensity when the light passed through alga liquids with different optical distances and different alga concn.were determined by using ST-85 automatic quantum illuminometer.[Result] As the alga cell concn.and optical distance were increased,the light intensity was decreased rapidly.When the optical distances were same,the light intensity was decreased along with the increase of alga cell concn.In primary culture period,when the alga cell concn.was lower,the light attenuation degree was less;in late culture period,as the alga cell concn.was increased,the light attenuation degree was increased gradually.The growth law of alga cell in culture process was similar to that of strains in microbial fermentation process.The biomass curve of alga cells was similar to the growth curve of strains in microbial fermentation.[Conclusion] The comprehensive influences of alga cell concn.and optical distance on light attenuation could be better described with Lambert-Beer equation:Ln(I/I0)=-KaXL.The growth curve of alga cells could be described with Logistic equation very well.
