摘 要:目的探索慢性肺动脉高压大鼠心脏结构和功能的变化。方法雄性成年SD大鼠40只,肺动脉高压组32只,采用腹腔注射野百合碱溶液建立慢性肺动脉高压模型;对照组8只,注射同样剂量的乙醇和生理盐水混合液。注射28d后通过右心导管测定肺动脉压力,评价造模效果。肺动脉高压组大鼠于建模前后行超声检测,测量反映左右心室结构和功能的指标。结果成功建立慢性肺动脉高压模型大鼠29只。肺动脉高压组肺动脉收缩压、肺动脉平均压和右室收缩压均升高,与对照组比较差异有统计学意义(P<0.01)。超声测量结果显示肺动脉高压组大鼠干预后舒张末期右心室上下径显著增大(P<0.05),左心室舒张末期横径及上下径变化不显著(P>0.05)。二尖瓣、三尖瓣E/A明显降低,与干预前相比差异有统计学意义(P<0.05)。左室射血分值、短轴缩短率及每搏量干预前后差异不具有显著性(P>0.05)。结论慢性肺动脉高压大鼠右室结构改变,左右心室的舒张功能下降,短期内左室收缩功能未发生明显改变。Objective To explore the characteristics of cardiac structure and function in a rat model with chronic monocrotaline-induced pulmonary hypertension.Methods The chronic pulmonary hypertension in rat model(n=32) was developed by monocrotaline solution injection into abdominal cavity,and 8 rats treated with ethanol and physiological saline were as control.The pulmonary artery systolic pressure was detected via the right heart catheterization,and the criterion of successful model was more than 30 mm Hg.The ultrasound test was taken to measure the structure and functional indicators of left and right ventricular in model rats before and after injection.Results Twenty nine models with monocrotaline-induced chronic pulmonary hypertension were successfully established.The length of right ventricular in end-diastolic diameter of pulmonary hypertension group significantly increased(P0.05).The pulmonary systolic pressure,pulmonary artery mean pressure and right ventricular systolic pressure accelerated apparently than that before monocrotaline solution injection(P0.01).The E/A of mitral,tricuspid significantly reduced(P0.05).Compared with the values before injection,there were no significant differences in LVEF,LVFS and SV after injection(P0.05).Conclusion The structural of right ventricular and the diastolic function of two ventriculars changed in the rat with chronic pulmonary hypertension.The systolic function of left ventricular does not decline significantly in short-term.
