摘 要:为解决马铃薯茎尖脱毒培养过程中黄化苗形成的问题,以马铃薯品种B01-31-9为试验材料,研究了茎尖诱导培养基中NH4NO3,MgSO4.7H2O,CaCl2.2H2O和铁盐浓度改变对茎尖培养过程中黄化苗发生及成苗率的影响。结果证明:培养基中氮、镁、铁浓度的改变对黄化苗的预防有显著作用,当NH4NO3,MgSO4.7H2O和铁盐的浓度分别在1 600 mg·L-1、400 mg·L-1、83.4 mg·L-1时,黄化苗发生率最低;但以上各因素处理对茎尖诱导分化成苗的影响差异并不显著。Experiments with different levels of NH4NO3,MgSO4.7H2O,CaCl2.2H2O and the iron salt in inducing medium were conducted by using cv.B01-31-9 in order to solve the problem of bud etiolation in potato shoot tip culture.Results showed that the concentration change of N,Mg and Fe had significant effects on etiolation inhibition,and the rate of etiolation was the lowest when the concentration of NH4NO3,MgSO4.7H2O and iron salt was 1 600 mg·L-1,400 mg·L-1,83.4 mg·L-1,respectively.However,there were no significant differences in generation index for all factors in etiolation experiments.
