摘  要:[目的]探索建立稳定的小鼠视神经损伤模型实验方法。[方法]选用成年C57BL/6小鼠60只,手术进行左眼视神经钳夹损伤,右眼设为正常对照眼,随机分为5个组,每组12只,分别在损伤后1d、3d、5d、7d、14d5个时间点摘除眼球。取眼球和视神经行冰冻切片,HE染色,观察组织形态并对神经胶质细胞计数。[结果]光镜下观察损伤部位形成损伤区域,损伤区域组织染色浅,未损伤眼视神经纤维排列整齐,胶质细胞与神经纤维走向一致,呈极性排列。损伤眼视神经纤维排列紊乱,胶质细胞杂乱无序。损伤后视神经胶质细胞计数减少,少于未损伤侧视神经,两者具有统计学差异。[结论]成功建立小鼠神经损伤模型,视神经损伤后形态学有显著改变。[Objective] To establish a stable animal model of optic nerve crush in mice.[Methods] The adult C57BL/6 mice(n = 60) were operated on left eyes to crush the optic nerve.The right eyes were set as controls.Mice were randomly di-vided into five groups according the sacrificed time at 1 d,3 d,5 d,7 d and 14 d after surgery.Eyeballs and optic nerves were gathered,and cryosectioned to observe and count the number of neuroglial cells by HE staining.[Results] The injured area was stained lightly.The uninjured optic neural fibers arranged neatly.The trend of the glial cells was as the same as the neural fibers.But in injured groups,the optic nerve fibers arranged in disorder and the glial cells lost polarity.The number of glial cells in injured optic nerve was significantly less than that in uninjured optic nerve.[Conclusion] The animal model of crushed optic nerve in mice was successfully established.The morphological alterations appeared in the injured optic nerve.
